In the twenty first century, supply chain operation has become the mainstream business model of enterprise. But now the probability of disruption risk is increasing, which may lead to huge economic losses. In recent years, with the improvement of people's living standards, the reinforcement of safety awareness, the advancement of science and technology and the large number of bio-pharmaceutical patents will soon expire, medical cold chain(MCC) industry is developing rapidly. Distribution center, as important link, location study has important significance, which determines the structure of the supply chain logistics network. This paper chose MCC logistics as research object and defined the scope of disruption risks, namely the function failure risk of distribution center. Then based on this, we explored distribution center location optimal problem.For the function interruption risk of distribution center, here a comprehensive analysis of internal and external risk factors had made, returning the occurring probability of interruption risks at different defensive levels and locations. Considering the particularity of the MCC, this paper combined the location strategy and inventory strategy with customer service strategy, then constructed distribution center multi-objective location optimization model of MCC under disruption risks. System location cost and service response level that compose.the location model optimization objective. Since the models are multi-objective NP-hard problems, the weighted TOPSIS and genetic algorithm were applied to solve them.By solving the model, we have the following decisions, involving location's total cost, the service response level, the location and defensive program of distribution center, the delivery and inventory strategies of MCC system. At the same time, via the comparative analysis of the results of numerical examples, considering disruption risk or not, sensitivity analysis of the key parameters of the model and the measurement of model objectives, we proved that the superiority of considering disruption risk and distribution center's fortify in the design stage. Meanwhile, according to their own actual conditions, the MCC managers can obtain more scientific decision proposals about location,defensive, delivery and inventory.
