Vancomycin and norvancomycin,which is a sugar peptide antibiotic,were prescribed by the ministry of agriculture to ban veterinary dr?gs.The ministry of health in 2011,issued the food may be illegal to add non-food substances and the abuse of food additives list(1-5 batch of summary)include vancomycin in announcement,and points out that “needs to develop animal foods was developed for the determination of vancomycin liquid chromatography-tandem mass spectrometry”.There is no standard for the detection of vancomycin and norvancomycin in animal derived foods.This paper establish a solid phase extraction-ultra high performance liquid chromatography-tandem mass spectrometry(UPLC-MS/MS)the determination of Vancomycin and norvancomycin in animal derived foods,inorder to fill the domestic blank detection method and ensure the safety of consumers.The following results were obtained:(1)A solid phase extraction ultra performance liquid chromatography tandem mass spectrometry(UPLC-MS/MS)method was developed for the determination of Vancomycin and norvancomycin in animal derived foods.The samples were extracted by organic solvent dichloromethane,removal of fat,protein and other impurities,purified with Strata-X-C SPE column,and were separated by ACQUITY UPLCTMBEH C18(50mm * 2.1 mm,1.7 ?g/kg)chromatographic column with acetonitrile and 0.1% formic acid-water(V/V)as mobile phase in gradient elution.Electron electrospray ionization(ESI)was used in positive ion multiple reaction monitoring(MRM)mode and quantified by external standard method.The results showed that Vancomycin and norvancomycin showed good linear relationship in 2?g/L~6000 ?g/L,the correlation coefficients were greater than 0.999.The detection limit(S/N=3)were 0.25 ?g/kg,0.40 ?g/kg,and the limit of quantification(S/N=10)were 1.0?g/kg,1.5?g/kg.The average recoveries of the two target compounds spiked at 2.5,10 and 25 ?g/kg,ranged from 80.4%~91.3%,with the relative standard deviations(RSD)less than 10%.The method can meet the requirements of rapid and accurate detection of Vancomycin and norvancomycin residues in animal derived food.(2)The statistical analysis of Vancomycin and norvancomycin residues in animal food in Guangdong Province was carried out.We conducted a random sampling of 189 batches of animal food,such as meat products,aquatic products,milk powder and milk,which were representative of Guangdong Province.The results showed thatthere were 2 batches of norvancomycin,including 1 batch of pork,the content of 5.9 ?g / kg and 1 batch of pig,the content of 10.2 ?g / kg,and the remaining 187 batches of samples were not detected Vancomycin and norvancomycin.The random sample data show that the province of animal food in general good,but still continue to increase supervision and enhance the quality of animal food safety.
