Due to excellent photo-physical properties of iridium complexes(large Stocks shifts,tunable UV-vis absorption spectra and fluorescence spectra by adjusting ligand,high quantum efficiency and long phosphorescence lifetime).Iridium complexes has attracted more and more attention in biological imaging and photodynamic therapy of tumors.However,the absorption of the traditional metal iridium complex is mainly located in the ultraviolet and visible region.Because of the small penetration depth of ultraviolet light and visible light,its application is limited in vivo.In this paper,it is extended to the near infrared region.The metal iridium complex and nanoparticles with near infrared absorption were successfully synthesized and their applications in photo acoustic imaging and photodynamic therapy were explored.The specific work includes the following two parts: 1.Synthesis and Properties of Fluorescence / Phosphorescence Dual Emission Metal Iridium ComplexesBecause of the strong absorption of cyanine dyes in the near infrared region and the photodynamic effects of the heavy atom iridium in the metal iridium complexes,we design and synthesize the near-infrared metal iridium complexs with the ligand of phenylpyridine and phenylquinolone.The photophysical properties and photostability of the metal iridium complexes were studied by UV-visible near-infrared absorption spectroscopy and emission spectra.The results showed that the iridium complexes had good photostability.2.Study on water-soluble metal iridium complex nanoparticles in photoacoustic imaging and photodynamic therapyIn order to improve the water solubility and biocompatibility of near-infrared metal iridium complexes,we selected a highly biocompatible polymer F-127,We make use of the hydrophobic hydrophobic interaction of the complex and the surfactant F-127 to make nanoparticles.The photophysical properties of the complex nanoparticles were studied by UV-visible-near-infrared absorption spectroscopy and emission spectroscopy.It was found that the complexes were encapsulated into nanoparticles,whose photostability was improved.Under the condition of near infrared laser irradiation,the nanoparticles have good photoacoustic imaging effect and singlet oxygen generation ability.The experiments in vivo show that the near infrared metal iridium complex nanoparticles have a good passive targeting effect on the tumor and can be used for photodynamic therapy guided by photoacoustic imaging.
