As the anti-cancer drug,taxol has attracted states widespread attention.The main source of taxol is from plant tissue extraction,yew cell culture,fully synthetic and semi-synthetic chemistry and microbial fermentation.Currently,taxol production mainly depends on the extraction in the yew bark,however,this production method due to the long period of tree growth,has been unable to make mass production.Therefore,search for its substitution and precursor substances instead of looking for itself,it is extremely necessary.Taxane and precursor substances are also continued to deepen.Endophytic fungi biosynthesis of taxol and taxane diterpenoids has its own advantages,and has been paid important attention by domestic and foreign researchers.In this study,used the taxol production strain Aspergillus niger subsp.taxi HD86-9 which has obtained as starting strain.Then fermentated it using bioreactors.After microbial fermentation,the fermentation broth and mycelium were extracted by ethyl acetate:methanol(V:V=1:1),then vacuumed distillation,finally got the extraction.Afterwards,utilizing normal phase chromatography,reverse phase chromatography,HPLC,recrystallization techniques,mass spectrometry and NMR,metabolites were isolated,purified and identified.Isolated four compounds in strain HD86-9 fermentation broth by 1H-NMR,13C-?MR,MS analysis,the test results show respectively 10-deacetyl-baccatinn,Baccatin ?,Taxol and Cephalomannine.This experiment carried endophytic fungi microbial fermentation metabolites which were isolated and purified and obtained biosynthesis precursors of taxol and made a foundation of transforming microorganisms obtained by chemical modification or paclitaxel,namely chemical semi-synthetic production of taxol.In addition,used the separation and purification technology in this experiment,the operation is simple,the separation effect is significant and the amount of solvent is small.Therefore,provides a useful reference for the separation and purification of paclitaxel and precursors.
