Chitosan is the basic polysaccharide in nature,but it widely attention in the field of medical biomedical materials because it safe non-toxic,and can be a variety of enzyme degradation characteristics in organisms.However,as a kind of polyelectrolyte,chitosan is not soluble in water,but in acid and little organic solvent due to it have a lot of hydrogen bond in intermolecular and intramolecular,limits the application.At present,how to keep the nature of the original chitosan,enhance its solubility in water seems to be one of the most hopeful ways to solve the problem.A novel soluble in water chitosan derivates,N-2-hydroxypropy dimethyl ammonium chloride chitosan(N-2-HFCC)was synthesized by the laboratory's homemade epoxy propyl methyl ethyl ammonium chloride grafted onto chitosan.In addition,Newcastle disease virus(NDV)La Sota capsulated in the N-2-HFCC/CMC nanoparticles(NDV/La Sota-N-2-HFCC/CMC-NPs)was prepared by the polyelec-trolyte complex method.The preparation technology of NDV/La Sota-N-2-HFCC/CMC-NPs was optimized by orthogonal test,the physicochemical characteristics,security,biological activity,release characteristics in vitro,and immune effect of NDV/La Sota-N-2-HFCC/CMC-NPs were systematic studied.The results of study demonstrate that the NDV/La Sota-N-2-HFCC/CMC-NPs produced by the optimal formulation have been produced with regular round shape,smooth surface,and well-distributed.The mean size of NDV/La Sota-N-2-HFCC/CMC-NPs was(252.2±32.68)nm,Zata Potential was(41.1 ±0.89)mV,entrapment efficiency was(96.6±0.72)%(n=5)and loading capacity was(53.2±1.11)%(n=5).The safety test were used to determine the of NDV/La Sota-N-2-HFCC/CMC-NPs was safety.Releasing tests in vitro has shown that NDV/La Sota-N-2-HFCC/CMC-NPs could make NDV sustained release.The results of stability test showed that NDV/La Sota-N-2-HFCC/CMC-NPs have good stability,and its biological activity did not change to save for three months in room temperature.Immune effect evaluation concluded that the NDV La Sota-N-2-HFCC/CMC-NPs can stimulate immune chickens to produce strong cellular immunity,humoral immunity and local mucosal immune.What's more,NDV La Sota-N-2-HFCC/CMC-NPs can have a stronger ability to resist of poison in SPF chicken,but also under strong poison attack show a better immune protection effect.This study constructed with N-2-HFCC/CMC as the carrier,Newcastle disease La Sota poison plant disease for mucosal immunity model drug delivery system,realized the NDV in the body for a long time,the purpose of and to improve local mucosal immunity and cell immunity,humoral immunity and enhance the immune effect of target.At the same time,provides the theoretical basis for the development of vaccine antigen delivery carrier,which is a safe,efficient and excellent performance.And this study has certain scientific significance and potential application prospects.
