In this paper;the research is focused on preparation a novel complex chitosan derivates sus'tained release microspheres.and on establishing a model with Newcastle disease' virus,based on contrarily electrostatic interaction process of chitosan derivatives.The size,polydispersity,zeta potential and morphology of the micro spheres were measured with transmission electron microscopy(TEM)and Zetasizer.Effect of N-2-hydroxypropyltimehyl ammonium chloride chitosan(HACC)concentration,carboxymethyl chitosan(CMC)concentration,NDV:HACC(v/v),the stirring speed and the stirring time on the mean diameter and drug entrapment efficiencies of drug-loading microspheres had been discussed.Optimization of the drug-loading micros pheres preparation conditions and study the drug release behaviour in vitro of the microspheres loaded with NDV.The results of study demonstrated that the drug-loading microspheres have been produced with suitable size,extremely spherical shape.The drug-loading microspheres produced by the optimal formulation were 304.3 nm of mean size,192.1-595.2 nm of size distribution and +32.50 mV of proper Zata Potential.The entrapment efficiency was(98.96±2.1)%.Furthermore,the results of releasing tests in vitro have shown that drug-loading microspheres could make NDV sustained release.Meanwhile,The delivery system of drug-loading microspheres had an excellent safety of being satisfactory,Which could be proved by cytotoxicity test.
