Electrochemical biosensor for the rapid,sensitive,low cost,convenient on-site detection,and has wide application prospect in food,chemical,pharmaceutical,clinical,biomedical and environmental analysis etc.Nano material has strong adsorption property and good biocompatibility,can well maintain the enzyme activity,and can enhance the electron transfer between the electrode and the enzyme active site,is an ideal material for biosensor construction.This paper combined with nanotechnology,materials preparation technology and other research areas,in the preparation of nano materials and nano materials application in biosensor development work,two kinds of biosensors were prepared to carry out specific work in the following two aspects:1.A new Voltammetric sensor for detecting Matrine was prepared,by dropping the graphene oxide coating dispensed onto bare glassy carbon electrode,use electrochemical reduction method reducing graphene oxide to ERGO after GO dried,then use the modified electrode to investigate of the electrochemical behavior of matrine.The results showed that,when detect matrine the constructed electrodes showed good sensitivity,stability and reproducibility.There exist the linear relation between the oxidation peak currents and matrine concentration in the range of 6×10-6~1×10-4mol L-1,with detection limit of 4×10-6.In addition,the electrodes exhibited better stability for application of the determination of matrine and provides the basis for routine analysis in matrine technology.2.In this experiment,the GO was reduced to rGO by direct electrochemical method,and then the HRP-CS/rGO-CS/GCE modified electrode was prepared.Because CS/rGO-CS can provide good conductive locations and a suitable microenvironment for HRP,and the composite film rGO has unique physical and chemical properties,which can effectively improve the HRP and the electrode surface electron transfer ability,so the modified electrode has ability of direct electrochemistry performance and catalytic oxidation reduction.Then the experimental conditions for the detection of hydrogen peroxide were optimized.Finally,the detection performance of hydrogen peroxide was investigated.When the concentration of H2O2 is in the range of 4×10-7~7×10-4mol/L,the response current is linear,and the detection limit is 1×10-7mol/L.The results show that the method is simple,low cost,good stability and high response sensitivity.It is a rapid and effective method for the determination of hydrogen peroxide.
