Paris polyphylla Smith var.chinensis(Franch.)Harais a rare and scarce Chinese herbal medicine in China.There are many medicinal properties,and the wild resources are extremely scarce because of excessive digging.Artificial cultivation can solve the shortage of resources.In artificial planting,seed reproduction is economical and efficient.However,in the course of germination,there are problems such as “Two Times Sleep”? low nursery stock?bring out seedlings untidy.Therefore,the solution of the Paris polyphylla Smith var.chinensis(Franch.)Hara seeds dormant has become a pressing problem.In this paper,the anatomical structure and the growth and development cycle of the seeds were observed.In this paper,the seed germination promotion of different temperature layer and three different concentrations of gibberellin were tested.The dynamic changes of three kinds of internal source hormones,ga,ABA and IAA,were determined in different stages of multilayered treatment,The results show:(1)The seed,which develops in natural condition,can be divided into five stages in the development cycle of Paris polyphylla Smith var.chinensis(Franch.)Hara embryo through the observation of the ordinary section and the paraffin section.The first stage: under natural conditions,the seed of the Huayuan building will appear when it grows 90 d,and the seed is a near circle with a longitudinal axis length of only 0.55 mm.The second stage: after 30 d,the embryo will grow to an ellipse,with the longitudinal axis of the embryo reaching 1.88 mm,which is 1/5 of the length of the longitudinal axis of the seed;Stage 3: after 45 d,the seed embryo length is 2.52 mm,1/3 of the longitudinal axis of the seed,ellipsoid;The fourth stage: after 40 d,the seed embryo length is 4.41 mm,which is 1/2 of the longitudinal axis of the seed.In the first four stages,the germplasm of the huazhuang mansion was not germinated.In the fifth stage,after 30 d growth,the Paris polyphylla Smith var.chinensis(Franch.)Hara germplasm can be completely broken through the skin.The first phase takes the longest time,and it takes three months for the building to grow.The next four stages take a short time.(2)The components of the seeds include: seed skin,embryo,germ,and milk.The longitudinal section is subglobose.The inside of the seed is full of ovule,and the milk cells are scattered in the form of spillage,which contains a large amount of amyloid.The cells at the center of the ovule and at the close patch of the germplasm are larger and are closely arranged and have a thick cell wall.The cells around the embryo are small and arranged more closely,and the wall is thin.The dermis is closely linked to the ovule.(3)The suitable reservoir conditions have a decisive influence on the seed germination of the Paris polyphylla Smith var.chinensis(Franch.)Hara.The ovules were immersed in 24 H with a concentration of 100 mg/l,followed by 45 d,4?,30 d,18-20?.It is the best condition for seed germination.In the fifth stage of the growth of the embryo at 150 d,the seed completely broke through the skin and the seed began to germinate.(4)The single gibberellin immersion treatment has no significant effect on the growth of the Warburg germplasm,and the variable temperature accumulation + gibberellin immersion treatment has certain influence on the growth of the warzao germline.The experimental results show that the most suitable concentration of gibberellin is 100 mg/l,and the high concentration of gibberellin(200mg/l)can inhibit the development of embryo.(5)Seed endogenous hormone has certain regularity in the growth process of germline.Ga content shows the tendency of rising and falling in the process of germination of huazhuang seeds.However,the changes in ga are not significant and are in a relatively stable state.The content of IAA has been on the rise until the embryo has completely broken through the skin.he change in IAA is more significant than that in ga.In short,the changes of ga and IAA content have a certain effect on the fourth and fifth stages of the growth of the embryo,and have little effect on the early stage of the growth of the embryo.This study can provide theoretical basis for seed germination morphology of the Paris polyphylla Smith var.chinensis(Franch.)Hara;To provide practical guidance for seed germination and artificial planting in the Paris polyphylla Smith var.chinensis(Franch.)Hara.
