Effects Of Electroacupuncture At Chongzhong, Sanyinjiao And Yinlingquan On Intravesical Pressure And ATP And MDA In Rabbits After Acute Urinary Retention

Acupuncture can be used to treat bladder diseases, such as enuresis, urinary retention, urinary incontinence. The point of Weizhong(BL40) is the Lower-He point of the bladder. According to the theory of "the Lower-He points are used to treat fu diseases", the point of Weizhong (BL40) was always the first choice to treat bladder diseases. This was recorded in many ancient books, but according to the national textbooks "Acupuncture And Moxibustion" and "The therapy of Acupuncture And Moxibustion", the point of Sanyinjiao(SP6) and Yinlingquan(SP9) are always chosen to treat bladder diseases first, while the point of Weizhong(BL40) is hardly mentioned. So it is necessary for us to explore the similarities and differences of these three points through experiments, and then to provide an important basis for the clinical choice.Objective:(1) To observe the effect of electro-acupuncture(EA) on the rabbits after acute urinary retention by needling the acupoints of Weizhong(BL40), Sanyinjiao(SP6) and Yinlingquan(SP9) respectively;(2) To find out which points are more closely connected with the bladder;(3) To explore the effective points on treating urinary bladder diseases.Methods:48healthy male adult rabbits were randomly divided into6groups (N=8):the normal group, the model group, the Weizhong (BL40) group, the Sanyinjiao (SP6) group, theYinlingquan (SP9) group and the non-acupoints group (the last four groups were considered as EA groups). In addition to the ones in the normal group, the other rabbits all had their bladder filled with saline which was2times to their normal bladder capacity for2hours to establish the acute urinary retention model. EA was used on the rabbits in the four EA groups right after they urinated. The EA was set with a frequency of2HZ/15HZ, a current intensity of0.6mA and with density waves. After20minutes, we tested their bladder pressure by a biological function signal acquisition system and then take their bladder tissue. After washing, weighing, homogenized, centrifuged, the bladder tissue were then placed and stored in the fridge at a temperature of-20?. The enzyme linked immunosorbent assay (ELISA) was used to test the contents of Adenosine Triphosphate (ATP) and Malondialdchydc (MDA) in their bladder tissue. At last, the experimental data were analysised by SPSS17.0statistical software.Results:(1) The bladder pressure:The bladder pressure of the rabbits reduced significantly after modeling, the difference was statistically significant (P<0.01). The bladder pressure of the rabbits in the EA groups increased significantly after EA treatment (P<0.05). After EA treatment, the increase rate of the bladder pressure of rabbits in the EA groups was higher than of the ones in the model group with a statistical significance (P<0.05). Comparing with the ones in the non-acupoints group, the rabbits in the other EA groups had more significant increase in their bladder pressure (P<0.05). But there were no significant differences (P>0.05) among the increase rate of bladder pressure the rabbits in the Weizhong (BL40) group, the Sanyinjiao (SP6) group and the Yinlingquan (SP9) group.(2) The ATP in the urinary bladder tissue: Comparing with the ones in the normal group, the contents of ATP in the urinary bladder tissue of the rabbits in the model group after modeling reduced significantly (P<0.05). Comparing with ones in the model group and in the non-acupoints group, the rabbits in the Weizhong (BL40) group, the Sanyinjiao (SP6) group and the Yinlingquan (SP9) group had significant increase in the contents of ATP in their bladder tissue after EA treatment(P<0.05). And after the EA treatment, there was still significant differences among the content of ATP in the bladder tissue in the Weizhong (BL40) group rabbits, the Sanyinjiao (SP6) group rabbits and the Yinlingquan (SP9) group (P<0.05) rabbits. The rabbits in the Weizhong (BL40) group had the biggest increase in the contents of ATP in their bladder tissue, and the Sanyinjiao (SP6) group took the second place. But there was no significant difference between the Sanyinjiao (SP6) group and the Yinlingquan (SP9) group (P>0.05).(3) The MDA in the urinary bladder tissue:Comparing with the ones in the normal group, the contents of MDA in the urinary bladder tissue of the rabbits in the model group increased significantly after modeling (P<0.05). Comparing with ones in the model group and in the non-acupoints group, the rabbits in the Weizhong (BL40) group, the Sanyinjiao (SP6) group and the Yinlingquan (SP9) group had significant reduction in the contents of MDA in their bladder tissue after EA treatment(P<0.05). Comparing with ones in the Yinlingquan (SP9) group, the rabbits in the Weizhong (BL40) group had higher contents of MDA in their bladder tissue after EA treatment(P<0.05), but no significant differences was found between the Sanyinjiao (SP6) group (P>0.05) and the other two groups.Conclusion:In this experiment, the bladder pressure, the content of the bladder tissue ATP and MDA were taken as the experiment indicator to observe the changing of the rabbits after EA. The results show that:(1) The bladder pressure of the rabbits decreased by50%after modeling, which shows that this modeling method is successful.(2) EA on the rabbits after acute urine retention by needling the points of Weizhong (BL40), Sanyinjiao (SP6) and Yinlingquan (SP9) can increase their bladder pressure, which means these three points can be used to treat acute urinary retention and bladder-related diseases.(3) EA treatment on the rabbits after acute urine retention by needling the points of Weizhong (BL40),Sanyinjiao (SP6) and Yinlingquan (SP9) can increase the contents of ATP in the bladder tissue, which means these three points are effective in treating acute urinary retention; The point of Weizhong (BL40) was most effective in increasing the contents of ATP in the bladder tissue, and the point of Sanyinjiao (SP6) took the second place.(4) EA treatment on the rabbits after acute urine retention by needling the points of Weizhong (BL40), Sanyinjiao (SP6) and Yinlingquan (SP9) can reduce the contents of MDA in the bladder tissue. The points of Weizhong (BL40) and Sanyinjiao (SP6) are more effective in reducing the contents of MDA in the bladder tissue than the point of Yinlingquan (SP9).
