Humans have accumulated rich of traditional knowledge of using plants to treat disease in their long-term practice,especially in some minority areas or indigenous communities in developing countries.China,a multi-ethnic populated country,possesses refulgent knowledge of ethno-medicinal plants.Unfortunately,the knowledge is eroding rapidly due to the ignorance of younger generations and the dying off of older generations who possess the knowledge.Therefore,it is extremely urgent to rescue this disappearing culture through modern ethnobotanical methodology.In this paper,traditional usage of plant has been surveyed by the method of ethnobotany in some ethnic minority areas of Guangxi and Guizhou.The results showed that Zhuang people not only used Tirpitzia sinensis(Hemsl.)Hallier for treatment of diminish inflammation,stop pain and bruises,but also used as handle of farm tool in Jingxi county,Guangxi.In addition,it has also been used as starter for preparing fermented beverages(especially local liquor)by Shui people in Sandu county,Guizhou.Moreover,Based on our literatures rearch,it has not been reported that the study on chemical component of T.sinensis by now.In this paper,T.sinensis,an ethnomedicinal plant in China,has been studied through the modern ethnobotanical approaches.Isolating and screening related pharmacological constituents would provide with scientific and reasonable evaluation for folk application of this plant,which will be benefit for inheritance and conservation the traditional knowledge of ethnomedicinal plant,and also can establish some theoretical basis on the plant's further development and application.Based on ethnobotanical survey and reviews of the studies on chemical constituents and its pharmacological activity of Linum plants,a group in the same family as Tirpitizia,we carried out the project on chemical constituents of T.sinensis.After 95%EtOH extraction,various column chromatography technologies,such as silica gel,MCI,macroporous resin,and Sephadex LH-20 chromatography,and instruments,including MPLC,HSCCC and Re-HPLC,were applied to isolate compounds from T.sinensis BuOH extract.Furthermore,modern spectrum technology(NMR and MS)and the methods of comparing with literatures were used to identify the structure of compounds.Twenty compounds has been determined to elucidate their structures,which were named(2S,3R)-methyl-7-hydroxy-2-(4-hydroxy-3-methoxyphenyl)-3-(hydroxymethyl)-2,3-dihydrobenzofuran-5-carboxylate(TS-20,1).Tirpitzoside A(TS-24,2).Tirpitzoside B(3.'-O-methylluteolin 6-C-[1"-?-glucoside-(6?1)-?-rhamnoside])(TS-25,3).Tirpitzoside C(TS-27,4).Adenosine(TS-28,5).3,5-Dimethoxy-benzyl alcohol 1-O-?-D-glucopyranoside(TS-33,6).Junipetriloside A(TS-35,7)and Leonuriside(TS-36,8),respectively.These compounds were firstly reported in Linaceae except compound 5,and 2,3 and 4 were new compounds.These results were not only important for chemotaxonomy of medicinal plants in the family Linaceae,but also contributed to its chemical constituents of T.sinensis.Meanwhile,it also enriched natural compounds for this species.According to the pharmacological activity of identified compounds,traditional usage of T.sinensis has been discussed,which will support its folk application.Besides,although the BuOH extraction is very hard to separating due to it contains plenty of strong polar component,this fraction usually be proved that have numerous significant biological active ingredients.Therefore,based on the experience in isolating BuOH extraction compounds,separation method of BuOH extraction was discussed,which would provide guidance for the separation of BuOH extraction.
