To Investigate the different mechanisms of Jingui Shenqi Pill Warming kidney yang and Liu Wei Dihuang Pill nourishing kidney yin from the Hypothalamic-Pituitary-Adrenal(HPA) Axis and Hypothalamic-PituitaryThyroid(HPT) Axis, and explore the differences mechanism of Reinforcing Kidney between the two prescription.MethodsTo establishment the Kidney yang deficiency mice model and the kidney yin deficiency model by Glucocorticoid, Observe the survival status of mice,Determination body mass of mice in each group, cyclic Adenosine Monophosphate(cAMP) content, cyclic Guanosine Monophosphate(cGMP) content by Enzyme linked Immunosorbent(ELISA) assay, Determination Adrenocorticotropic Hormone(ACTH) content, Cortisol(Cor.) content in HPA Axis, Thyroid Stimulating hormone(TSH) content, Triiodothyronine(T3) content, Thyroxine(T4) content in HPT Axis by Radioimmunoassay, calculate the Adrenal and Thyroid index in each group, and observed the morphological changes in Adrenal and Thyroid tissue.Results1. Gastric perfusion with 25 mg / kg HI for 10 consecutive days can result a Kidney yang deficiency mice model, Kidney yang deficiency model group mice appears some symptoms,such as mental fatigue, reduced activity, unresponsive, toe end of the cold, twist arch, fur dull, anorexia, loose stool. With prolonged administration, the symptoms gradually worsened, Body weight decreased(P<0.01), Plasma cAMP content increased(P<0.01), Plasma cAMP content decreased(P<0.01).3. Jingui Shenqi Pill could increase Serum ACTH, Cor. content in HPAAxis, and Serum TSH,T3,T4 Content in HPT Axis(P<0.01), increase Adrenal and Thyroid index(P<0.05),and repair damaged Adrenal and Thyroid tissue.4. Liu Wei dihuang Pill could decrease Serum ACTH, Cor. content in HPAAxis,and Serum TSH,T3,T4 Content in HPT Axis(P<0.01), increase Adrenal and Thyroid index(P<0.05),and repair damaged Adrenal and Thyroid tissue.ConclusionThe change of Plasma cAMP and Plasma cAMP content could be one of the important indicators of Investigation glucocorticoid Kidney yang deficiency model and Kidney yin deficiency model can be successful.
