Objective:Based on the speculating,association,abstraction,purification and observation to human life phenomenon,the TCM theory of Qi was established Gradually by ancient Chinese people,and then the theory of TCM Vitality was formed inch by inch.The needs for treatment and health care guided by the theory of TCM Vitality become more and more intense.However,the connotation of TCM Vitality has been fuzzy and cannot be measured.So,further development and clinical application of the theory of TCM Vitality in the modern social environment are hindered to a certain extent.In this article,the relationship between Hypothalamic-Pituitary-Thyroid Axis and TCM Vitality will be analysed,the similarity between deficiency syndrome of TCM Vitality and nonthyroidal illness syndrome?NTIS?will be cogitated.Aimed at investigating how to create deficiency syndrome of TCM Vitality animal models,observing the changes on Hypothalamic-Pituitary-Thyroid Axis and TCM Vitality in SD rat after right kidney excision,exploring the appropriate indexes for measurement of TCM Vitality objectively and researching the mechanism of Vitality consolidation Treatment.Method:1.Theoretical study:Collect and learn ancient and modern documents on Hypothalamic-Pituitary-Thyroid Axis,TCM Vitality and animal model of TCM Syndrome,analyse the relationship between Hypothalamic-Pituitary-Thyroid Axis and TCM Vitality,cogitate the similarity between deficiency syndrome of TCM Vitality and NTIS,seek the method for create animal model of Vitality deficiency.2.Experimental study:The right kidney of 8 weeks old SD rat was removed,then the traditional Chinese medicine Nourishing strength Da Bu Yuan Jian was given to rats by gavage since the third week after operation.The hypothalamic pituitary thyroid axis was taken as a starting point.The general state of rats was observed as well as their weight and ear temperature.Autonomous behavioral activity detection and water maze experiment were used to detect the activity,the learning and memory ability and the exploration consciousness of the rats.Hormones of the hypothalamic-pituitary-Thyroid Axis and liver deiodinated enzyme activity were tested by Enzyme-linked immunosorbent assay.Morphological features of thyroid was observed by HE dying.Expression of mRNA in type 1 and type 3 deiodinated enzyme genes of liver and genes associated with Hypothalamus pituitary thyroid axis functional regulation were detected by Real Time-PCR,as Expression of protein by Western Blot.Results:1.The general state?ear temperature and weight of rats:?1?After the right nephrectomy,the most rats curled,flocked together,arched the back,cut down their activities and the hair became withered and yellow.Part of the rats was in a fret with diarrhea,stinky fart,shallow and fast breathing even accompanied by asthma and increase in the amount of urine.While there was no obvious abnormality in the blank control group.?2?There was no obvious change in the ear temperature of the rats in the blank group at each time point.In the first week after the operation,the rats in model group,high dose of Da Bu Yuan Jian group?abbreviated as DG group?,Dabu Yuan Jian middle dose group?abbreviated as DZ group?and low dose group of Da Bu Yuan Jian group?abbreviated as DD group?rat ear temperature higher than those in control group,but the difference was not statistically significant.In the second week after operation,ear temperature of model group,DG group,DZ group and DD group rat was lower than that in control group,but the difference was not statistically significant.In the third week after operation,compared with model group,ear temperature of DG group,DZ group and DD group rat have rebounded,but lower than that of the control group;compared with the control group,the ear temperature of model group rats decreased?P<0.05?;comparison with the model group,DZ group rat ear temperature increased?P<0.05?.In the sixth week after operation,compared with the control group,the ear temperature of model group rats decreased?P<0.05?;compared with the model group,ear temperature of DZ group rat increased?P<0.05?.?3?There was no significant difference in weight gain in one week of rats in each group before operation and in the first week after operation?P>0.05?.In the third,fifth week after operation,the weight gain of the rats DZ group was lower than that in the blank group and the model group?P<0.05?.In the seventh week after operation,compared with the blank group,the weight gain of the rats in the model group,DG,DZ and DD groups were significantly lower than that in the control group?P<0.01?.2.Behavioral tests:?1?autonomic activities detection after 6 weeks of gavage,compared with the control group,the total distance,average speed,rest time,central time of the model group rats was respective significantly shorter,slower,longer,shorter?P<0.01?.Compared with the model group,the average speed of DG group rats was faster?P<0.05?,and its'rest time was significantly shorter?P<0.01?;the total distance of DZ group rats was shorter?P<0.05?,and its'average speed was faster?P<0.05?;the rest time of DD group rats was significantly shorter?P<0.01?,and its'central time was longer?P<0.05?.?2?Morris water maze test showed that,compared with the control group,the swimming distance of the rats in the model group was significantly longer?P<0.01?,and its'average speed was significantly slower?P<0.01?,spend more time to find the platform?P<0.01?;Compared with the model group,the swimming distance of DG group rats was significantly shorter?P<0.01?,the average speed was significantly faster?P<0.01?,spend significantly shorter time to find the platform?P<0.01?;DZ group rats'swimming distance was significantly shorter?P<0.01?,the average speed is faster?P<0.05?,spend significantly shorter time to find the platform?P<0.01?;DD group rats'swimming distance was significantly shorter?P<0.01?,spend significantly shorter time to find the platform?P<0.01?.?3?Spatial exploration test showed that,compared with the blank group,model group rats'number of traversing platforms was fewer?P<0.01?,and its'time in the target quadrant was shorter?P<0.01?.Compared with the model group,rats in DZ group arrived platforms more times?P<0.05?,DG group rats stayed in the target quadrant longer?P<0.05?,DZ group rats stayed in the target quadrant significantly longer?P<0.01?.3.Hypothalamus-pituitary-thyroid axis hormone detection:compared with blank group,serum FT4 and T4 levels of model group rats were significantly lower?P<0.01?,T3 and TSH lower?P<0.05?,while serum rT3 level was significantly higher?P<0.01?.Compared with the model group,serum FT4 levels of three drug groups rats were significantly higher?P<0.01?,serum rT3 levels were significantly lower?P<0.01?;serum T3 level of DZ and DD groups rats were higher?P<0.05?.4.Real Time-PCR test showed that:compared with the control group,TRH mRNA from the hypothalamus of the model group rats,TRHR mRNA from hypophysis,TSHR mRNA from thyroid showed a down-regulated expression;compared with the model group,DZ group rats'TRH mRNA was up-regulated,and three drug delivery groups rats'TRHr mRNA expression were up-regulated in different degree,DZ group rats'TSHR mRNA was up-regulated.Compared with the control group,the model group rats'TRH protein level in hypothalamus,TRHR protein level in hypophysis decreased,and TSHR protein level in thyroid increased;compared with the model group,DG group rats'TRHR protein level in pituitary,TSHR protein level in thyroid increased,DZ and DD group rats'TRHR protein level in pituitary increased,TSHR protein level in thyroid decreased.5.Liver D1,D3mRNA expression and the activity of type 1,type3deiodinated enzyme:compared with the control group,the model group rats'liver D1 mRNA showed a down-regulated expression,liver D3mRNA showed a down-regulated expression,the activity of liver type 1deiodinated enzyme decreased?P<0.01?,the liver activity of type 3deiodinated enzyme increased,but the difference was not statistically significant?P>0.05?.Compared with the model group,DG?DZ and DD groups rats'liver D1 mRNA showed a up-regulated expression,DG?DZ and DD group rats'liver D3 mRNA showed a down-regulated expression;the activity of liver type 1 deiodinated enzyme in DZ,DD groups increased?P<0.05?,the activity of liver type 3 deiodinated enzyme in three drug groups decreased,but the difference was not statistically significant?P>0.05?.Conclusion:1.The function of TCM Vitality is similar to that of hypothalamic pituitary thyroid?HPT?axis.2.The performance of rats after right nephrectomy is similar to that of syndrome of deficiency of TCM Vitality.3.Da Bu Yuan Jian has a positive effect on rats with right nephrectomy,and the mechanism may be affecting the hypothalamic pituitary thyroid axis.
