| Macleaya Cordata(Willd.) R. Br. is a plant of Papaveraceae family which is widely distributed in the south of the Yangtze River and the north of Nanling, also it exhibits good activities such as anti-inflammatory, insecticidal function, regulating animal intestinal flora,promoting animal growth and so on. Macleaya Cordata mainly contains alkalids,besides it contains flavonoid, terpenoids, steroids, polysaccharides and so on. In this study, the mass spectrometry tracer and modern separation techniques are adopting to separate the chemical compounds of the leaves in Macleaya Cordata, finally 17 compounds are isolated which are identified by IR, ESI-MS, 1H-NMR, 13C-NMR, 2D-NMR, etc.1 previous researchBased on the early analysis of HPLC-Q-TOF/MS in the leaves of Macleaya Cordata,the results showed that there are many species of alkaloids among which are undetected,2 Extraction and separation of chemical constituentsFresh leaves of Maleaya Cordata were divided into three parts: acid precipitation part,alkali precipitation part, alkaline liquor part after shattering, extracting with 95% ethanol and the method of alkali extraction and acid precipitation. Then the alkaline liquor part was adopting as the sample through the analysis of TIC.Totally 17 compounds were isolated using column chromatography and semi-micro preparative liquid chromatograph combined with mass spectrometry tracer technique, among which one compound was new and three were the first found in Macleaya Cordata.3 Identification of the compoundThe sructure of 13 compounds were identified through chemical and spectral method,which were dihydrochelerythrine, 6-Cyan-dihydrosanguinarine,(1’→6)-hydroxyethyl dihydrosanguinarine, dihydrosanguinarine,(1’→6)-hydroxyethyldihydrochelerythrine,6-acetonyldihydrochelerythrine, sanguinarine, chelerythrine, 6-acetonyldihydrosanguinarine,tetrahydrocolumbamine, cheilanthifoline, carboxymethyldihydrochelerythrine,4’-hydroxy-5’-methoxy-cinnamenyl-(1’→6)-dihydrosanguinarine, among which4’-hydroxy-5’-methoxy-cinnamenyl-(1’→6)-dihydrosanguinarine was a new compound andcarboxymethyldihydrochelerythrine, tetrahydrocolumbamine, cheilanthifoline were the first time found in Macleaya Cordata. |
