Abstract/Summary:
Objective:Study Liuwei dihuang decoction on D-galactose rapid aging mice cognitive function and bFGF/Notch signal path expression, and explore the possible mechanism of liuwei dihuang decoction to improve learning and memory capacity.Methods:Mice were randomly divided into normal group, model group, the six flavors of high, middle, low dose group,Vit E group, D-galactose subcutaneously injected method is used to copy the rapid aging mice model,at the same time to the corresponding intervention, 8 lines of nerve function score over the weekend, with Morris water maze system to determine the change of the learning and memory of rats, mice brain after using immunohistochemical method and rt-pcr method to detect small non-colored bFGF, Notch1, Hes1 on the number of positive cells and the expression of protein levels.Results:(1) body quality and weight: compared with normal group, model group and treatment group is apparent neurologic deficits symptoms, highly significant difference(P<0.01); Compared with model group, the treatment group of nerve function defect symptoms(P<0.01); Within each treatment group, the six flavors high dose group compared with other treatment group have statistical significance(P<0.05), the effect is best.(2) in mice after building lengthen the incubation period, the collapse of fewer(P<0.05), while the treatment group compared with model group,the incubation period shorten, the collapse of number increase(P<0.05),liuwei dihuang decoction high dose group and low dose group, Vit E group comparison, the incubation period is short, fall more frequently, with significant difference statistically significant(P<0.01);(3) compared with normal group, model group bFGF, Notch1, Hes1 positive cells was less, highly significant difference(P < 0.01), the treatment group of bFGF expression, Notch1, Hes1 positive cell number were higher than in model group(P<0.01), the six flavors high dose group of bFGF, Notch1, Hes1 positive cells was significantly higher than that of other groups, highly statistical significance(P<0.01)(4) compared with normal group, model group bFGF, Notch1, Hes1 positive cells was less, the treatment group of bFGF, Notch1, Hes1 m RNA expression were higher than in normal group and model group(P<0.01),The six flavors high dose group of bFGF, Notch1, Hes1 m RNA is significantly higher than other groups, highly statistical significance(P<0.01).Conclusion:Liuwei dihuang decoction can improve the D- galactose rapid aging mice learning memory disorders; Increase the expression of bFGF in aging mouse, activation of Notch signaling pathways, may be the mechanism of liuwei dihuang decoction anti-aging brain puzzle.
