Objective1.To study the effect of alcohol concentration in the blood of Wuzhiyin in rats.2. To study the effect of Wuzhiyin on the liver of rats with acute alcoholism 3.To observe the protective effect of Wuzhiyin on acute alcoholic liver injury and gastric mucosalinjury. Methods1. The effect of alcohol concentration in the blood of Wuzhiyin in rats:40 SD rats were randomly divided into model group, low dose group of Wuzhiyin, high dose group of Wuzhiyin, and Gehua group. Prophylactic to 30 min after the administration of medicine, each in accordance with the 1.5m L/100 g wine(alcohol 52%) given by gavage, drunk 1h, 2h, 3h, the inner canthus vein plexus puncture blood, detected the blood ethanol concentration by GC(gas chromatography)2.The effect of Wuzhiyin on the liver of rats with acute alcoholism:50 SD rats were divided into 5 groups: blank group, model group, Gehua group, low dose group and high dose group with Wuzhiyin. After preventive administration, the blank group distilled water was poured into the stomach, and the remaining groups were treated with 1.5m L/100g(alcohol degree 52%) for 1 weeks, and the model of acute alcoholic liver injury was established.The content of serum alanine aminotransferase(ALT), aspartate aminotransferase(AST) were detected; The content of liver homogenate glutathione(GSH), superoxide dismutase(SOD) and malondialdehyde(MDA), alcohol dehydrogenase(ADH) were detected.3.The effect of Wuzhiyin on acute alcoholic liver injury and gastric mucosalinjury:After the rat blood collection,to collect liver and stomach as samples, and observe the effect of Wuzhiyin on the alcohol induced liver injury in rats. Results1.Compared with the model group, both low dose group of Wuzhiyin and high dose group of Wuzhiyin can reduce the alcohol concentration of blood(P < 0.05, P < 0.01), The concentration of ethanol in blood in each group:high dose group of Wuzhiyin< low dose group of Wuzhiyin< Gehua group < model group.In this experiment, the chromatographic method was linear with good(correlation coefficient R=0.9995), good precision(RSD=1.41%), the sample was stable in 96h(RSD=4.63%),recovery rate of 90.10%~107.85%(RSD=5.19%).2. Wuzhiyin can reduce ALT,AST activity, reduce the generation of GSH, so that the MDA content increased, improve the SOD, ADH activity, compared with the model group has statistical significance(P<0.05), and high dose effect is more obvious.3.Wuzhiyin in high and low dose groupcan reduce alcohol induced liver injury.The effect of Wuzhiyin on the liver of rats with acute alcoholism The gastric mucosal lesionindex of low dose group of Wuzhiyin was lower than that of model group(P<0.05), and the high dose group of Wuzhiyin was also lower than thatof model group(P<0.01). The results of HE staining showed that Wuzhiyin could reduce the damage of alcohol induced liverinjury and gastric mucosal injury, and the effect of high dose group of Wuzhiyin was more obvious. Conclusion1.Wuzhiyin can reduce the blood concentration of ethanol and high dose decreased more significantly.2.Wuzhiyin with hepatoprotective effect and its mechanism of action may promote alcohol decomposition and the antioxidation effects of reduced liver injury through activation of ADH.3.Wuzhiyin canimprove the acute alcoholic liver injury, gastric mucosal injury, show protective effect on stomach.
