Objective: The expressions of leptin and leptin receptor in papillary thyroid carcinoma and in nomrmal thyroid tissues in nodular goiter tissues are evaluated to see whether there are statistical differences between the three. And then the relationship between its expression and clinical and pathological features of patients is analyzed to provide the basis for the clinical prognosis of papillary thyroid carcinoma patients.Methods: 63 pathological sections of thyroid cancer operation patients hospitalized in department of emergency general surgery of Affiliated Hospital of Qingdao University from October 2012 to December 2013 are selected, One Have other organs cancer ? two conducted chemoradiotherapy are removed. 60 cancer tissues are remained as thyroid cancer group, from which thyroid normal tissues are chosen as Normal group. Another 60 tissues which affirmed is nodular goiter tissues worked as nodular goiter groups. Immunohistochemistry(non-biotinylated two-step method) is applied to detect the expression of leptin and leptin receptor in the sections of the three groups and their differences are found. Then according to the clinical indicators including papillary thyroid cancer patient's age, gender, tumor size,depth of invasion, lymph node metastasis and BMI, comparisons are made between the groups and the expressions of leptin and receptor are analyzed to discuss the relationship of leptin and leptin receptor with the clinical and pathological features of patients with papillary thyroid cancer.Results:(1)In papillary thyroid cancer group, there are 46 cases of positive expression of leptin, 14 cases of negative expression, with the positive expression rate of 76.67%. In normal group there are 32 cases of positive expression of leptin, 28 cases of negative expression, with the positive expression rate of 53.33%. In C group there are 30 cases of positive expression of leptin, 30 cases of negative expression, with the positive expression rate of 50.00%.Both have significant statistical differences(P <0.05).(2)45 cases show positive expression of leptin receptor in papillary thyroid cancer group and 15 cases show negative expression, with the positive expression rate of 75.00%. 36 cases indicate positive expression of leptin receptor in B group and 24 cases have negative expression, with the positive expression rate of 60%. 33 cases indicate positive expression of leptin receptor in C group and 27 cases have negative expression, with the positive expression rate of 55%. There are statistical differences between the two groups(P <0.05).(3)The expressions of leptin and leptin receptor in papillary thyroid cancer are significantly increased, indicating that leptin system may be associated with the occurrence of papillary thyroid cancer.Further study on the information of papillary thyroid cancer patients reveals that the positive expression rates of leptin and its receptor in age(?45)are higher than those in age(<45), the positive expression rates with lymph node metastasis group are higher than those without lymph node metastasis group, all of which have statistical differences. However in the group comparison on tumor size, depth of invasion and BMI, the expressions of leptin and its receptor have no significant differences statistically.Conclusion: leptin and its receptor is obviously correlated with the occurrence and development of papillary thyroid cancer. It may become an indicator to determine the prognosis of patients with papillary thyroid cancer.
