Effect Of Qi Li Jie Qing Fang On The Expression Of TNF-?, IL-1? And IL-6 MRNA In Rats With Gouty Arthritis

Objective : By intraperitoneal injection of potassium oxonate combined with Coderre prepared by the improved model of gouty arthritis,the level of influence of Juanbilijieqingfang on the expression of inflammatory cytokines in synovial tissue in a rat model of TNF-alpha,IL-1 beta,IL-6 gene,to evaluate its anti gouty arthritis with and provide mechanism.The experimental and theoretical basis for the clinical application of the treatment of gouty arthritis.Methods : Select the SPF grade healthy 96 male SD rats were randomly divided into normal control group,model group,Juanbilijieqingfang group,etoricoxib group;after modeling,etoricoxib group given 11mg/kg intragastric administration of Juanbilijieqingfang group using concentrated liquid 44g/kg gavage the above 4 groups,each daily gavage 2 times,for 2 days,observe the general condition of the rats,ankle swelling index,serum uric acid concentration and pathological changes of synovial tissue,and the pain threshold by semi quantitative RT-PCR method to detect the synovial tissue of TNF-alpha,IL-1 beta,IL-6 mRNA expression changes.Results :1.After the success of the model,compared with the control group,the rats in model group hair in different degree of dry,yellow,shedding,eating,drinking water is reduced,the subjects of ankle swelling,skin temperature increased,the anatomy of uric acid crystals visible pigmentation,ankle joint swelling index and hematuria acid level increased significantly(P<0.01).Joint swelling index(P<0.01),changes under the light microscope showed obvious pathology of synovial tissue,that improved gouty arthritis model was successfully established;compared with the model group,Juanbilijieqingfang group and drug intervention group at each time point after the swelling index decreased(P<0.05),the changes of pain threshold(tail reaction time extension)(P<0.01),decreased serum uric acid content(P<0.01),visible synovialcell hyperplasia and infiltration of inflammatory cells in light microscope,small vessels decreased,TNF-alpha,IL-1 beta,IL-6 mRNA Decreased expression(P<0.05).2.By semi quantitative RT-PCR assay,Juanbilijieqingfang group,relying on examination of tumor necrosis factor alpha in synovial tissue group(TNF-alpha),interleukin 1 beta(IL-1 beta),interleukin 6(IL-6)compared with that in the model of gene expression decreased(P<0.05);Juanbilijieqingfang group and no significant difference between group,no statistical significance(P>0.05).Conclusion :1.The modified rat model of gouty arthritis was successfully reconstructed by continuous intraperitoneal injection of potassium and Coderre.2.Juan-Bi-Li-Jie-Qing-Fang has anti-inflammatory and analgesic,good for gouty arthritis rats decreased serum uric acid,further proof of the validity,its mechanism may be related to its inhibition of rat ankle joint synovitis factor TNF-alpha,IL-1beta,IL-6 release.
