Objective:Through the experiment,the establishment Gegenqinlian colonic localization tablet(GGQLJC,Gegenqinlian colonic localization tablet)method for the determination and HPLC fingerprint active ingredients,the main peaks preliminary herbs home,and anti-rabbit Damp type UC preparation to study the mechanism and provide theoretical basis for controlling the quality of the preparation GGQLJC and explore its therapeutic mechanism damp type UC.Method:1.Using high performance liquid chromatography,Agilent ZORBAX SB-C18(4.6×250mm,5?m)column was used with acetonitrile(B)-0.015mol ·L-1 potassium dihydrogen phosphate solution as the mobile phase gradient elution,detection wavelength:270nm;flow rate:1mL · min-1;column temperature:30?;injection volume:10?L.Detection time:120min.Using fingerprint similarity calculation software(2004A Edition)for data analysis,GGQLJC HPLC fingerprint chromatograms according to the retention time and the chromatographic peak corresponding to GGQLJC fingerprints main peaks preliminary herbs home,while in the compound determine the content of the active ingredient.2.The use of high fat and high carbohydrate diet made the rabbit Damp type physique,combined with trinitrobenzene sulfonic acid(TNBS)was established Damp type UC model;Rabbits were divided into seven groups of eight:normal group,model group,GGQLJC high dose group,GGQLJC dose group,GGQLJC low dose group,positive medicine sulfasalazine(SASP)group,traditional Gegenqinlian contiguous(GGQL)group;clinical activity of rabbits observation,DAI score,CMDI score,TDI score,measuring the length of the colon,spleen index,thymus index and the index of the colon,detected by ELISA in serum and colon tissue MPO and Th1/Th2 cytokine levels by immunohistochemical staining colon tissue PPAR-y and NF-kBp65 protein expression,the treatment groups were compared to explore GGQLJC treatment damp type UC pharmacodynamic mechanism and immunological mechanisms.Results:1.Adoption of the methodological study to establish a qualitative and quantitative detection methods GGQLJC fingerprint,and fingerprint detection and analysis conducted 12 batches Gegenqinlian colon positioning tablets were detected a total of 35 peaks,of which 13 peak attributed to Kudzu,13 a peak attributed to a skullcap,six peak attributed to berberine,four peak attributed to licorice;similarity of 12 batches of the formulation were greater than 0.9,the production preparation process is stable.2.Pharmacodynamic Experimental Studies showed that the treatment group can effectively alleviate the clinical symptoms of UC Damp type rabbits,reducing DAI scores,CMDI score,TDI score,increasing the length of the colon,reduce colon and spleen weight,increase the weight of thymus reduce MPO activity,decreased rabbit serum and colon tissue Thl cytokines IL-1? and TNF-? and increase Th2 cytokines IL-4,IL-13 expression,the expression of PPAR-y increases colon tissue,reduced colon tissue NF-kBp65 protein expression compared with model group were significantly different(P<0.01),which GGQLJC chip high-dose group and SASP group effect is more significant.Conclusion:HPLC chromatographic methods established effective quality control GGQLJC and GGQLJC can improve Damp type UC inflammation,regulate the balance of protein protein expression Thl/Th2 cells and PPAR-y and NF-kBp65,its effect is better than traditional GGQL group for the study of traditional Chinese medicine oral colon delivery system has laid a certain experimental basis.
