Objective: We explored the repair mechanism underlying the protective effect of moxibustion on gastric mucosal lesions by investigating the level of endogenous protective factor such as epidermal growth factor(EGF),nitric oxide(NO)and Tumor Necrosis Factor ?(TNF-?)in rat gastric mucosal injury lesion after the operation of nucleus of solitary tract(NTS),to supply experimental basis for moxibustion in the treatment of gastric mucosal lesions in clinical application.Methods: 48 SPF grade healthy SD rats were twice randomly divided according to the random number table: Normal group(Group A,n=12),Model group(Group B and C,n=24),NTS damaged group(Group D,n=12).Then,divided Model group into Model group(Group B,n=12)and Moxibustion group(Group C,n=12).Firstly,Three days of recovery after bilateral NTS damaged in Group D,while others were fed normally.Secondly,Group A was given saline and another 3 groups were administered of absolute ethanol at dose of 6 ml/ kg and aspirin at dose of 200 mg / kg for consecutive three days to establish gastric mucosa injury model.After that,the moxibustion was applied on Zusanli(ST36)except to the group A and group B,which was done twice a day and lasted for 3 days.Finally,we adopted the serum and gastric mucosa to observe the gastric biopsy ulcerindex(UI)and detect the level of EGF,NO and TNF-? by Enzymes Linked Immunosorbent Assay(ELISA).Results: 1.The gastric mucosal injury index(UI)with the level of EGF and TNF-? in tissue as well as the TNF-? in serum had obviously increased(P<0.01 or P<0.05)while the level of NO in serum and tissue was significantly decreased(P<0.01)in group B,C,and D compared with group A After gastric mucosal injury and moxibustion stimulation,Which had varified the success of model making.2.The gastric mucosal injury index(UI)with the level of TNF-? in serum and tissue was obviously decreased(P<0.01)while the level of EGF and NO in serum and tissue were significantly increased(P<0.01)after moxibustion stimulation in group C compared with group B.It indicated that moxibustion stimulation could regulate the content of endogenous protective factors and promote the repair of injured gastric mucosa.While the gastric mucosal injury index(UI)and the level of EGF NO and TNF-? in serum and tissue in group D changed a little(P? 0.05)and without statistical significance.3.The gastric mucosal injury index(UI)and the level of TNF-? in serum and tissue were increased(P<0.05 or P<0.01)while the level of EGF and NO in serum and tissue were decreased(P<0.05 or P<0.01)after NTS damaged in group D compared with group C.It explained that it was unconspicuous to the effect of moxibustion on gastric mucosa repair because of the blocked of nerve pathway after NTS damaged.Conclusion 1.Moxibustion can promote the repair of damaged gastric mucosa by regulate the content of EGF,NO and TNF-in serum and tissues.2.The effect of moxibustion on gastric mucosa repair was unconspicuous after NTS damaged,which was explained that the nucleus of the solitary tract is an important part of the signal pathway in the repair of gastric mucosal injury.
