Objective:To select 5 kinds of traditional Chinese medicine Alpinia?Galangal,Alpinia galanga Swartz,Fructus alpiniae galangae,Alpinia katsumadai,Alpinia oxyphylla?petroleum ether,ethyl acetate,n-butyl alcoho and water area as the research object,establish the model of gastric ulcer with cold syndrome rats on the 5 kinds of traditional Chinese drugs resistance in four parts,through observing the effects on behavior rat signs,cyclic nucleotide system,ATP enzyme,sympathetic adrenal system,endocrine system,summed up the common medicinal function foundation,analysis the internal relationship may exist,for the study of traditional Chinese medicine.The material basis of relationship based on the provided experimental evidence.Methods:By gavage of glacial acetic acid and 4?rhizoma anemarrhenae Decoction to establish a rat model of cold syndrome of gastric ulcer.After the success of modeling,were given the blank group and the model group were fed with normal temperature,positive group were fed with ginger,cimetidine,water soluble compounds,40 tested drugs group were fed with corresponding drugs,every 12 h administered1 times,a total of 4 times administration.To observe the changes of rat behavior characterization?appearance,behavior,weight,rectal temperature,tongue,wool color score,toe color changes,stool,mental state?,using enzyme detection of gastric tissue cAMP?c GMP?AC?PDE?Na+-K+-ATP?Ca2+-Mg2+-ATP and blood serum ACTH?D-?-H?T3?T4?TSH content ELISA,and calculate the ratio of cAMP/cGMP.Results:1.The effects on the behavior of ratsCompared with the blank group,model group rats stool half congealed dilute,listlessness,slow;compared with the model group,the subjects of drug rat stool volume decreased,relatively dry,forming mental state has been restored,the action is more flexible.Compared with the blank group,model group,rectal temperature and weight were significantly decreased?P<0.05?,wool color score was significantly increased?P<0.01?;each group of petroleum ether and ethyl acetate in high dose group anal increased rectal temperature?P<0.05 or P<0.01?,each group of petroleum ether and ethyl acetate in high dose group?except galanga ethyl acetate group?increased weight?P<0.05 or P<0.01?,each group of petroleum ether,ethyl acetate,n-butanol and water fraction of the high dose group and low dose group?except Alpinia oxyphylla petroleum ether,Alpinia galanga Swartz n-butanol,Alpinia katsumadai n-butanol,Alpinia galanga Swartz water,Fructus alpiniae galangae water,Alpinia oxyphylla water fraction of the low water fraction extracts high dose weight increased?P<0.05?.There were no significant differences in the anal temperature,weight,wool color score in the other polar regions.2.The effects of cyclic nucleotide system on rats gastric tissueCompared with the control group,model group cAMP,the ratio of cAMP/cGMP decreased significantly?P<0.01?,the PDE values was significantly increased?P<0.01?,the AC values were significantly decreased?P<0.05?;all groups had no significant difference in cGMP value.Each group of petroleum ether and ethyl acetate in high dose group AC values,cAMP value,cAMP/cGMP ratio were significantly increased?P<0.05 or P<0.01?,PDE decreased?P<0.05 or P<0.01?;and Fructus alpiniae galangae,Alpinia katsumadai,Alpinia oxyphylla n-butanol fraction in high dose group cAMP increased?P<0.05?,Fructus alpiniae galangae,Alpinia oxyphylla n-butanol fraction in high dose group increased cAMP/cGMP ratio?P<0.05?,Alpinia galanga Swartz water in high dose group increased AC?P<0.05?,Alpinia oxyphylla n-butanol fraction in high dose group decreased PDE?P<0.05?.The rest of the polar fraction were AC values,PDE value,cAMP value,cAMP/cGMP ratio did not change ether and ethyl acetate in high dose group?except Alpinia katsumadai petroleum ether,Alpinia oxyphylla ethyl acetate group puzzle?Na+-K+-ATP increased?P<0.05 or P<0.01?,each group of petroleum ether increased?P<0.05 or P<0.01?;Alpinia galanga Swartz water fraction,Fructus alpiniae galangae n-butanol fraction in high dose group and Fructus alpiniae galangae,Alpinia katsumadai,Alpinia oxyphylla petroleum benzin fraction,Galangal,Alpinia katsumadai,Alpinia oxyphylla ethyl acetate fraction in low dose group ACTH increased?P<0.05?,Fructus alpiniae galangae n-butanol fraction and water fraction in high dose group,Fructus alpiniae galangae petroleum ether fraction,Galangal,Alpinia katsumadai ethyl acetate fraction in low dose group D-?-H increased?P<0.05?.There was no significant difference in the ACTH value and D-?-H value of other polar parts.5.The effects of endocrine system in ratsCompared with the blank group,model group,T3 value,T4 value,TSH value decreased significantly?P<0.01?;each group of petroleum ether and ethyl acetate in high dose group?except petroleum ether fraction of Alpinia galanga Swartz group?T3 increased?P<0.05 or P<0.01?,each group of petroleum ether ethyl acetate,high dosage group of T4 value and TSH value were increased?P<0.05 or P<0.01?;Galangal,Alpinia katsumadai n-butanol fraction in high dose group was significantly T3 value ratsCompared with the blank group,model group large area of gastric mucosal injury,obvious exudation,necrosis,with mucous layer,muscular layer granulation tissue hyperplasia and a large number of inflammatory cell infiltration;Each group of petroleum ether and ethyl acetate fraction in high dose group gastric mucosal lesion area was significantly reduced,exudation,necrosis,the infiltration of inflammatory cells was significantly reduced,mucous layer,muscular layer hyperplasia of granulation tissue increased significantly,each group of n-butanol,water fraction in high dose group of gastric mucosal injury area decreased,exudation,necrosis and infiltration of inflammatory cells reduced,mucosa layer,muscle layer hyperplasia of granulation tissue increased;The changes of gastric mucosal injury,inflammatory cell infiltration and granulation tissue hyperplasia were not obvious in the other polar parts.Conclusion:5 kinds of Alpinia herbal medicine has improve the behavior representation,improve the model of gastric ulcer with cold syndrome rats regulate cyclic nucleotide system function and can improve the ATP activity,enhanced sympathetic adrenal system and thyroid function playing warm and cold dispelling effect,protect gastric mucosa,and playing the material basis of improve temperature blood,warming stomach for dispelling cold effect mainly in the petroleum ether fraction and ethyl acetate fraction in the minority,the traditional Chinese medicine has affinity,there is a correlation between the resistance and material basis.
