Objective Bushen Shengxue Jiedu Fang(BSSXJDF)is a Chinese herbal decoction.We aimed to investigate the molecular mechanisms of this herbal decoction on the expressions of T cell immune response related factor;the restored dynamic balance of T cell immune response network system;the appreciation and differentiation of hemopoietic stem cells(HSCs)and the rebuilt hematopoietic function of bone marrow.Methods Our animal model of mice,except mice in normal group,received a sublethal total body irradiation of 60Co?-irradiator and immune-mediated Cedicine.Drug treatment was started began after lymph node cell infusion,and lasted for 28 days.Mice were randomly divided into four groups as follows:normal group,model group,Cyclosporin(CsA)group as positive contrast,BSSXJDF group.We observe the effects of hematology indicators.after treatment.With immunofluorescent labeling,we using a laser confocal scanning microscope to observe the expression and distribution of T-bet from T lymphocyte,and the combination of SAP and SLAM.We use flow cytometry instrument to detect the number change of T?B?NK cells from peripheral blood and spleen.We use flow cytometry instrument to detect the proportion change of Th1?Th2?Th17?Treg cells.We use flow cytometry instrument to detect apoptosis rate of bone marrow hematopoietic stem cells.Results(1)On the 7th?14th day after treatment,the HGB and RBC counts of all treatment groups doesn't change much,as compared with the model group(P>0.05).On the 21th?28th day after treatment,the HGB and RBC counts of all treatment groups proliferate significantly,as compared with the model group(P<0.01).On the 7th day after treatment,the PLT counts of all treatment groups doesn't change much,as compared with the model group(P>0.05).On the 14th?21th?28th days after treatment,the PLT counts of all treatment groups proliferate significantly,as compared with the model group(P<0.01).After the molding,the WBC counts of normal group proliferate significantly,as compared with the model group(P<0.01).On the 21th days after treatment,the WBC counts of all treatment groups proliferate significantly,as compared with the model group(P<0.01).When the all treatment have been done,the change of RDW?MCV?MCH?MCHC is exist,as compared with the model group(P<0.05),but lack of regular.(2)The results of immunofluorescence show that,the T-bet counts from T cell of normal group proliferate significantly,as compared with the model group(P<0.01).The T-bet counts of all treatment groups drop significantly,as compared with the model group(P<0.01).The combined rate of SAP and SLAM of normal group drop significantly,as compared with the model group(P<0.01).The combined rate of CsA group proliferate significantly,as compared with the model group(P<0.01).The combined rate of BSSXJDF group proliferate slightly,as compared with the model group(P<0.05).(3)The results of cells change in peripheral blood,who detected by flow cytometry instrument,show that the rate of T cell in peripheral blood of normal group drop significantly,as compared with the model group(P<0.01).The T cell rate of all treatment groups drop significantly,as compared with the model group(P<0.01).The rate of B cell in peripheral blood of normal group does't change much,as compared with the model group(P>0.05).The rate of NK cell in peripheral blood of normal group proliferate significantly,as compared with the model group(P<0.01).The rate of B?NK cell of all treatment groups drop significantly,as compared with the model group(P<0.01).The rate of Th2?Th17 cell change without statistical significance(P>0.05).The Th1?Treg cell rate of normal group proliferate significantly,as compared with the model group(P<0.01).The Thl?Treg cell rate of all treatment groups drop significantly,as compared with the model group(P<0.01).The results of cells change in spleen,show that the rate of T cell in spleen of normal group drop significantly,as compared with the model group(P<0.01).The T cell rate in spleen of all treatment groups drop significantly,as compared with the model group(P<0.01).The rate of B cell of normal group change without statistical significance,as compared with the model group(P>0.05).The B cell rate in spleen of all treatment groups drop significantly,as compared with the model group(P<0.01).The NK cell rate in spleen of normal groups proliferate significantly,as compared with the model group(P<0.01).The NK cell rate in spleen of BSSXJDF group drop significantly,as compared with the model group(P<0.01),CsA group has downward trend,as compared with the model group(P<0.05).The Thl cell rate of normal group proliferate significantly,as compared with the model group(P<0.01).The Thl cell rate in spleen of BSSXJDF group drop significantly,as compared with the model group(P<0.01).The Th2 cell rate of molding groups drop significantly,as compared with the normal group(P<0.01).The Th2 cell rate of all treatment groups change without statistical significance,as compared with the model group(P>0.05).The Treg cell rate in spleen of normal group drop significantly,as compared with the model group(P<0.01).The Treg cell rate in spleen of BSSXJDF group proliferate slightly but without statistical significance,as compared with the model group(P>0.05).The Thl7 cell rate of normal group change without statistical significance,as compared with the model group(P>0.05),but all treatment groups drop significantly,as compared with the model group(P<0.01).The results of apoptosis in bone marrow,show that the rate of apoptosis of normal group proliferate significantly,as compared with the model group(P<0.01).The apoptosis rate of all treatment groups drop significantly,as compared with the model group(P<0.01).Conclusions Our data indicate that BSSXJDF,a traditional chinese herbal decoction,could treat aplastic anemia disease through regulating T cell immune response network,restoring dynamic balance of T cell immune response network in bone marrow,and regulating the appreciation and differentiation of HSCs.
