Objects:Purifying flavonoids from the head inflorescences of Coreopsis tinctoria and determine their structures.Studying contents of main ingredients in medicinal materials and the part of flavonoids.Studying traits of different batches of Coreopsis tinctoria.Analyzing the relationship of contents of main ingredients and traits of Coreopsis tinctoria.To finding active ingredients in flavonoids through pharmacological experiment and lay the foundation for the quality standard of Coreopsis tinctoria.Methods:Using Silica gel?Sephadex LH-20?RP-18 and Semi-PHPLC with the help of recrystallization to purify flavonoids from the extract of Coreopsis tinctoria.Using 1H-NMR?13C-NMR?HMQC?MS to determine structures of flavonoids.Using HPLC-DAD to determinate contents of main ingredients in medicinal materials and the part of flavonoids.Building chromatograms of Coreopsis tinctoria and evaluating their similarity.The color characteristics of the flower were observed,and counting the color depth and quantitative character of reddish brown.Ten batches of samples are grouped with cluster analysis performed by SAS verison 9.4.The relationship between the content of the main flavonoids and the color of flowers are analyzed.The effects of flavonoids on the glucose metabolism of 3T3-L1 preadipocytes are studied by glucose oxidase method.The dosages are 2.5,5,10?g·mL-1.Combined with chemical,traits and pharmacological studies to clarify the relationship between the contents of flavonoids and the quality of medicinal materials,lay the foundation for the establishment of quality standards.Results:There are 12 compounds isolated and 8 compounds in those identified.They are 2S-3',5',5-trihydroxyflavone-7-O-?-D-glucopyranoside,3',5'-dihydroxyflavone-7-O-?-D-glucopyranoside,2S-3',5,5 ',7-tetrahydroxyflavone,2S-3',4',7,8-tetrahydroxyflavone,marein,okanin,iguano-4'-O-?-D-glucopyranoside,quercetin-7-O-?-D-glucopyranoside.4 major components' content in the part of flavonoids are determined.The results show that the content of 2S-3',4',7,8-tetrahydroxyflavone is 0.05371 mg mg-1,the content of 2S-3',5',5-trihydroxy dihydroflavone-7-?-D-glucopyranoside is 0.032 12 mg·mg-1,the content of marein is 0.118 8 mg·mg-1,the content of okanin is 0.047 37 mg mg-1.According to the similarity analysis,it is found that the similarity of HPLC samples are 0.95 plus.The result of traits study showed that there are differences in the traits.The results also showed that the content of marein and okanin in different batches of samples was different.The content of marein was positively correlated with the color and area of reddish brown part.The results of pharmacological studies showed that the part of flavonoids and components of the head inflorescences of Coreopsis tinctoria could affect the glucose metabolism of normal mice 3T3-L1 cells,but there was no significant difference between the test group and the control group.Conclusion:The flavonoids in head inflorescences of Coreopsis tinctoria mainly contain chalcone and dihydroflavone,and the highest content of marein is related to the traits.It is suggested that marein should be used as the index component of quality evaluation.
