ObjectiveThe global incidence of high blood pressure is rising,and the high incidence increases the burden on individuals,families and society.Elevated blood pressure can cause damage to the heart,brain and kidney of target organs,cause vascular remodeling,and may be associated with systemic metabolic changes.Without early detection and appropriate treatment,it can lead to myocardial infarction(MI),stroke,renal failure,and death.In addition,hypertension can promote the development of other diseases.In the real society,various stressors stimulate the incidence of hypertension.JNC-7 first proposed the concept of pre-hypertension.The purpose is to reduce the incidence of hypertension.And it is consistent with the concept"prevention before disease onset" of traditional Chinese medicine.Based on this,we prepared the model of stress-induced pre-hypertension(SI-PHT)rats by foot-shocks combined with noise stimulation,intervened with the twirling-reducing(TR)needling method to investigate thoroughly the mechanism of TR needling method on SI-PHT rats from the genetic perspective.Methods36 Wistar male rats were randomly divided into blank control,model control and TR needling intervention groups(n=12 per group).The SI-PHT rat models except those in blank control group were prepared by combining electric foot-shocks with generated noise.Molding cycle lasted for 11 days.And TR needling stimulation on bilateral Taichong(LR3)and Quchi(LI11)was applied on TR needling intervention group during model preparation.The systolic blood pressure(SBP)and behavior change of rats were recorded the day before intervention and on the 3rd,5th,7th,9th and 11th day after modeling.After 11 days,gene expression in hypothalamus of each group was analyzed based on gene chip technology.ResultsAscending range of SBP in three groups was less than 20mmHg.SBP in blank control group stayed normal during the whole process of the experiment(P>0.05).SBP in model control and TR needling intervention groups whose behavior changed inordinately like mania elevated significantly(P<0.01)compared with that of rats in blank control group from the 3rd to the 11th day of the experiment.SBP in TR needling intervention group increased remarkablylr<0·01)compared with that of rats in model control group from the 5th to the 11th day of the experiment.Meanwhile,93 genes were upregulated and 153 were downregulated in model control group compared with blank control group(|FC|>1.5,P<0.05).59 genes were upregulated and 86 were downregulated in TR needling intervention group compared with model control group(|FC|>1.5,P<0.05).And 40 genes expressed adversely between model control versus blank control group and TR needling intervention versus model control(|FC|>1.5,P<0.05).Among the 40 genes,Acsm3,Cga,CryaB and Tsh? are closely related to hypertension(|FC|>1.5,P<0.05).TR needling intervention lowered blood pressure via activating the autoimmune thyroid disease(AITD)pathway.ConclusionBy acupuncture irritability prehypertension rats with bilateral Taichong(LR3)and Quchi(LI11)point,TR needling method show the anti-hypertensive effect on model rats;Under the same conditions,the risk of developing hypertension for SI-PHT rats with TR needling method intervention is lower than that of the model control group.TR needling method can cause SI-PHT rat hypothalamus hypertension related gene Acsm3,Cga,CryaB,Tsh? differentially expressed,the possible biological mechanisms for the differentially expressed genes including biological process and molecular function changes,but the booster mechanism during the building(model control group compared with the blank control group)and acupuncture anti-hypertensive mechanism of TR needling method(TR needling method group compared with model control group)may be different.During the process of blood pressure increasing,TR needling method showed the anti-hypertensive effect via AITD pathway;its possible machanism is that thyroid hormone levels rised causing vascular resistance down,resulting in blood pressure ascensional range slower in TR needling group compared with model control group increased blood pressure process.It is unclear wheather the insulin resistance mechanism associated with hypertension is activated while TR needling method showed the anti-hypertensive effect via AITD pathway.Further research will be done.
