Objective:Hypertension is a common disease,is mainly manifested as increased systemic arterial blood pressure,is unclear in most patients.Essential hypertension is a type of hypertension,accounting for more than 95%of patients with hypertension disease.EH The main clinical manifestations of EH are often contact with heart,brain,kidney and other target organ functional and(or)organic disease.These days the gene chip rapidly developing a biotechnology that uses for test and analysis of gene expression as appropriate to explore multigene-induced of hypertension.In this experiment,we used gene chip technology and RT-PCR technology to investigate effects of acupuncture on Renying(ST9)acupoint on the gene expression of hypothalamus in spontaneous hypertensive rats(SHR)and analyze internal mechanism to provide a new idea for acupuncture treatment of Essential hypertension.Methods:We randomly divided 20 SHR into Renying(ST9)and model control groups,10 body weight-matched Wistar-Kyoto rats were used as blank control group.All the rats in three groups were loosely immobilized in a specially made restrainer,the acupuncture method was applied to Renying(ST9)group,which is needling Renying(ST 9)acupoint on both side of the neck,for 19 min and hand-manipulating of needle for 1 min,every 6 days,1 day rest,total 28 days of needling.Before and after acupuncture treatment the 1,5,9,13,17,21,25 and 28 days measured variety of systolic blood pressure.The model group and normal group received no treatment except immobilized in the restrainer.The intervention was last for 28 days,4weeks and after Rat Gene 2.0 array technology was used for determination of gene expression profiles,and the key genes were verified by RT-PCR technology.Results:1.Measured results of blood preassure:to compare model group and normal group at 1,5,9,13,17,21,25 and 28 days was increased;to compare model group and Renying group on 1st 5th 9th days systolic blood pressure was reduce(P<0.01).The model group on 13,17,21,25,28 days blood preassure was increased(P<0.05).2.Gene Chip results:The different groups exhibited differential gene expression:compared with the blank control group,48 genes were up regulated and 91 genes were down-regulated in the model group;with the model group,79 genes were up-regulated and 80 genes were down regulated in Renying(ST9)group.3.The RT-PCR results of the key genes including 5-HT1A and CBS were consistent with gene chip analysis(fold-change?1.5).Conclusion:Acupuncture at Renying(ST9)acupoint can significantly reduce the blood pressure of SHRs and affect the hypothalamus gene expression profile.The principle of anti-hypertension in acupuncture may be related to the expression of CBS and 5-HT1A genes associated with neurotransmitters and the excitability of sympathetic nerve.
