Objective9-month-old female senescence-accelerated mouse prone 8(SAMP 8)model mice were selected as the research objects.To observe the learning and memory ability and central neurotransmitter levels and discuss the relationship between them by electroac upuncture(EA)at Sanyinjiao(SP6)of SAMP8 female mice,for providing experimenta 1 evidence for clinical treatment.Method9-month-old SAMP8 mice(number 12)were randomly divided into two groups:model model group and EA group,6 in each group.At the same time,9-month-old S AMR1 mice were the control group(number 6).Fixed the mice,EA treatment group was accupunctured at "Sanyinjiao(SP6)"acupoint;the control group and model contro 1 group were no EA treatment at the same time.2/100Hz were given,20 min at a ti me,every each other day,for 15 times.Morris water maze examination was used to detect the learning and memory ability in mice;The hippocampus tissues were collect ed for detecting the contents of acetylcholine(ACH)?acetylcholinesterase(ACHE)and Cholin Acetyltransferase(CHAT)by enzyme-linked immunosorbent assay(ELISA).The monoamine neurotransmitter tissues were collected for detecting the contents of Dopam ine(DA)?Norepinephrine(NE)?5-hydroxytryptamine(5-HT)by High-performance liqui d chromatography-electrochemical detection(HPLC-ECD).SPSS statistical software 20.0 was used for analysis.Result1.Morris water maze experiment1.1 Place navigationCompared with the control group,the escape latency of the model group were prol onged and there were statistically differences(P<0.01);the escape latency of the EA g roup were prolonged.From the first day to the fourth day,there were statistically diff erence(P<0.05,P<0.01)and in the fifth day,there was no difference(P>0.05).Comp ared with the model group,the escape latency of the EA group was shorter than that of the model group and there was statistically difference(P<0.01,P<0.05).1.2 Spatial probeCompared with the control group,the numbers mice crossing of model group obv iously decreased and the difference was statistically difference(P<0.01);the numbers mice crossing of EA group decreased and there was no statistical significance(P>0.05)Compared with the model group,the numbers mice crossing of EA group was longer and there was statistically difference(P<0.01).Compared with the control group,the time spent in target quadrant of the model group was shorter than that of the control group and there was statistically difference(P<0.01);the time spent in target quadrant of the EA group was longer than that of t he control group and there was no difference(P>0.05).Compared with the model gro up,the time spent in target quadrant of the model group was longer and there was st atistically difference(P<0.01).Compared with the control group,the percentage of the distance spent in target q uadrant of model group was lower than that in the control group and there was statist ically difference(P<0.01);the percentage of the distance spent in target quadrant of m odel group was higher than that of the control group and there was no difference(P>0.05).Compared with the model group,the percentage of the distance spent in target quadrant of EA group was prolonged and there was statistically difference(P<0.01).2.Comparison of cholinergic neurotransmitters in the hippocampusAcetylcholinesterase(AChE):Compared with the control group,the AChE content in hippocampus of model group was decreased and there was statistically difference(P<0.01);the AChE content of EA group was decreased and there was no difference(P>0.05).Compared with the model group,the AChE content of EA group was higher than that of the model group and there was statistically difference(P<0.05).Choline acetyltransferase(ChAT):Compared with the control group,the content o f ChAT in hippocampus of model group and the EA group were decreased and there was statistically difference(P<0.01).Compared with the model group,the ChAT conte nt of EA group was higher than that of the model group and there was statistically di fference(P<0.01).Acetylcholine(ACh):Compared with the control group,the ACh content in hippo campus of model group was decreased and there was statistically difference(P<0.01);the ACh content of EA group was decreased and there was no difference(P>0.05).C ompared with the model group,the ACh content of EA group was higher than that of the model group and there was statistically difference(P<0.05).3.Comparison of monoamine neurotransmitters in the cortex5-hydroxytryptamine(5-HT):Compared with the control group,the 5-HT content i nhippocampus of model group was decreased and there was statistically difference(P<0.01);the 5-HT content of EA group was decreased and there was no difference(P>0.05).Compared with the model group,the 5-HT content of EA group was higher than t hat of the model group and there was statistically difference(P<0.01).Norepinephrine(NE):Compared with the control group,the NE content in hippoc ampus of model group was decreased and there was statistically difference(P<0.05);t he NE content of EA group was decreased and the difference was no difference(P>0.05).Compared with the model group,the NE content of EA group was higher than th at of the model group and there was no statistically difference(P>0.05).Dopamine(DA):Compared with the control group,the content of DA in hippoca mpus of model group was decreased and there was statistically difference(P<0.01);th e DA content of EA group was decreased and there was statistically difference(P<0.05).Compared with the model group,the DA content of EA group was higher than th at of the model group and there was no difference(P>0.05).ConclusionEA at "Sanyinjiao(SP6)" may have improvement on the learning and memory ab ility of SAMP8 mice through influence on the process of ACh production in the hipp ocampus and monoamine neurotransmitters in the cortex.
