| Objective:Perimenopausal syndrome which refers to a series of syndromes in perimenopausal women or thereafter due to the recession of the ovarian function and declining estrogen levels, giving priority to disturbed vegetative neural symptoms, such as hot flashes, night sweats, depression, anxiety and so on. Perimenopause is an important turning point in women’s physiological changes, it is highly concerned for it can cause a series of physical, psychological and social problems. With the multiplication of social pressure and the growing of more and more aged population, irregular lifestyle,the tension in the family,there will be varying degrees of mental symptoms such as anxiety, depression,and it will turn into perimenopause anxiety when the symptoms become serious.With the improvement of women’s cultural knowledge level,to undertake more and more pressure of society, career, family, The incidence rate of the disease shows rising tendency, and affect the perimenopausal women’s physical and mental health, at the same time leave a great burden to family and society.And the study has found its occurrence is closely related to the disorders of the central nervous system neurotransmitter.Western medicine treatment generally take estrogen replacement therapy (hormonal replacement therapy, HRT) as the first approach to slow the ovarian senescence, and to achieve the purpose of prevention and treatment of perimenopausal syndrome.However, recently more and more reports about the varieties of side effects of alternative therapies cause people begin to seek more safe and effective methods of treatment. Acupuncture as one of TCM therapies, playing an important role in perimenopausal syndrome.It not only have the exact curative effect, but also less adverse reaction. For the predisposing factors and different symptoms in women’ perimenopausal syndrome we can choose the best prescription based on differentioation of syndrome,It gets more and more international attention. but the clinical selection of acupoints mainly based on the subjective judgment of the doctors, and the applications of acupoints are complicated and changeable, so there is no objective criteria and basis on clinical work.Previous research has shown only acupuncture effect results, but not discuss the function and mechanism in the middle of the process.It is generally believed that the center at all levels of the brain regulate reproductive endocrine function, Receive the incoming information of its internal and external environment, After integrating, the controlled information is passed to each function system by autonomic nerves and neuroendocrine system.The changes of the central nervous system to perimenopausal syndrome may include the abnormal expression of the central neurotransmitter and receptor, and the excitability of cerebral functional area, etc.Neurotransmitters as the intermediated material between NIM network and perimenopausal syndrome has the irreplaceable important role.The experiment is on the basis of preliminary work, in the past we selected many animal models ovariectomized rats.This experiment choose rats model with natural menopause as the objects of observation. By the therapy of acupuncture, We observe the impact of acupuncturing(EA) Sanyinjiao Acupoint on natural menopause rats in the aspects of central monoamine neurotransmitters in hippocampus and the rats’ anxious behavior changes in order to further to explain the role and the mechanism of neurotransmitter in the perimenopausal anxiety,so as to offer scientific basis for acupuncture treatment of endocrine disorders in perimenopausal women.Methods:The clean female rats were divided into the normal control group, the model group and the treatment group. The rats in treatment group were treated with electro-acupuncture at Sanyinjiao acu-point once every other day for15times therapies. The rats in the other two groups were raised with conventional methods, and only will be caught and stimulated every other day.Results:1. The rat behavioral test results for each groupThe elevated plus-maze test: Compared with the normal group, OE%, OT%in rats of model group significantly decreased, HD values significantly increased,which showed obvious condition of anxiety (with P<0.05or P<0.01).Compared with the model group, OE%, OT%in rats in the treatment group significantly increased, HD values significantly decreased (with P<0.05or P<0.01), but for the values of OE+CE and RE, there is no statistically significance (with P>0.05).2. The comparing results of the content of E2in rats ovary in each groupCompared with the normal group, the content of E2in rats’ovaries in model group significantly reduced (with P<0.01), Compared with the model group, the content of E2in rats ovaries’in the treatment group significantly increased.3. The comparing results of the content of the brain monoamine neurotransmitter in each groupCompared with the normal group, the content of5-HT of hippocampus in the model group inceased abnormally, and the ratio of5-HT/NE significantly increased, but the content of NE, DA decreased significantly (with P<0.01).Compared with the model group, the content of5-HT and the ratio of5-HT/NE in the hippocampus of the rats with the treatment group significantly deceased, the difference was statistically significant (with P<0.01or P<0.05), and the content of NE significantly increased (with P<0.05), though DA content increased but has no statistically significance (with P>0.05).Conclusion:1. EA"Sanyinjiao"point can intervene the appearance of anxiety symptoms during perimenopause, it showed better anti-anxiety effect.2. EA "Sanyinjiao" point can adjust central monoamine neurotransmitter levels in naturally menopausal rats, adjust autonomic nerve function and improve the menopausal rats behavior symptoms.3. Acupuncture can adjust the content of monoamine neurotransmitters to change the imbalance of the NIM network, thus caused the beneficial changes in estrogen levels. |
