ObjectiveThe purpose of this study based on the theory of "lift off" medicine for drug laws,under the guidance of fructus viticis and inula flowers as the carrier,verify the lifting and medicinal effects on IPF angiogenesis in rats,furthermore,to explore the core role of apoptosis in angiogenesis,and clear the pathogenesis of pulmonary fibrosis.Methods(1)Literature review:By retrieving access to the CNKI,ten thousand party and VIP database keyword to "lift off","pulmonary fibrosis","apoptosis","angiogenesis" clinical reports,retrieve the fixed number of year for 2000-2018,application study of the theory of lifting off,and effect of cell apoptosis,angiogenesis and its significance in idiopathic pulmonary fibrosis process analysis system to sort out.According to the in pulmonary fibrosis disease,apoptosis and angiogenesis play a role in pulmonary fibrosis disease,combining the theory of lifting and medicinal,observation of fructus viticis,inula flower apoptosis of pulmonary fibrosis rats and the influence of the degree of angiogenesis,provide theoretical basis for the application of traditional Chinese medicine to improve pulmonary fibrosis.Literature review:by retrieving access to the CNKI,ten thousand party and VIP database keyword to "lift off","lifting medicine for","herbal","pulmonary fibrosis","apoptosis","angiogenesis" clinical reports,retrieve the fixed number of year for 2000-2018,application study of the theory of lifting off,and effect of cell apoptosis,angiogenesis and its significance in idiopathic pulmonary fibrosis process analysis system to sort out.According to the in pulmonary fibrosis disease,apoptosis and angiogenesis play a role in pulmonary fibrosis disease,combining the theory of lifting and medicinal,observation of fructus viticis,inula flower apoptosis of pulmonary fibrosis rats and the influence of the degree of angiogenesis,provide theoretical basis for the application of traditional Chinese medicine to improve pulmonary fibrosis.(2)Experimental study:Experiment using SPF Wistar rats,male,80,in weight(190-220)g,randomly divided into 5 groups,were divided into control group,model group,fructus viticis,inula flower group,dexamethasone group.the rest of the group adopt lai is called trachea drip into the method to establish model of pulmonary fibrosis in rats.? Histopathologic action research:Geting lung tissue after molding 7d,28d,and HE dyeing,under a microscope the pathological pattern in rat lung tissue,alkaline solution is used to detect the fresh expression of hydroxyproline content in lung tissue.? Cells apoptosis research:taking alveolar lavage fluid after molding 7d,28d,and extracting alveolar macrophages,flow cytometry instrument testing each rat alveolar cell apoptosis rate changes.? Angiogenesis research:taking alveolar lavage fluid,the rats serum and lung tissue after molding 7d,28d,ELISA method to detect alveolar lavage fluid,MIP-la/MCP-1 the expression level,ELISA method to detect bFGF and PF4 expression level in serum.VIII related antigen measuring pulmonary microvascular density after dyeing,after HE dyeing pulmonary arteriolar wall thickness measurement.Results(1)Literature review:The theory of lift off is one of the important contents of traditional Chinese medicine theory.Useing this theory to treat disease,can be better guidance on clinical medication.Lung,the officer of assistant,for control,mainly related to gas,breathing,the main dispersing pertains to the descending.Treatment of pulmonary disease lung lift in coordinating,adopt xuan and shi,opening and closing method is given.The flower is rose,inula flowers fall alone;All school,fructus viticis alone.Both can into the lung,fructus viticis uplink releasing lungs,inula transforming lung qi,phlegm drink.Combination of fructus viticis and inula medicinal and from the Angle of angiogenesis and cell apoptosis carding process of pulmonary fibrosis disease mechanism,it is concluded that the pathogenesis of pulmonary fibrosis for "apoptosis as the forerunner,angiogenesis is more significant".(2)Experimental study? Histopathological studyIn the IPF in rats induced by bleomycin model,HE dyeing result shows inflammatory infiltration in lung tissue area and pathological changes of fibrosis area is very obvious,on the basis of alveolitis and pulmonary fibrosis severity,to carry on the classification rate,compared with control group,each group made the mold after all has the very significant difference(P<0.05),after dosing 7d and 28d,compared with model group,fructus viticis,inula flower with significant difference(P<0.05).Rats after building,model group HYP expression quantity increases,compared with the control group with significant difference(P<0.01);7d and 28d when the expression of HYP content in lung tissue dosage group were significantly lower than the model group,the difference compared with model group significantly(P<0.01).Building 7d,inula flower expression of HYP content is higher than in m group,compared with ground rice group with significant difference(P<0.01),fructus viticis and ground rice group differences had no statistical significance;Building 28 days,fructus viticis expression of HYP content is higher than in m group,with significant difference between the m group comparison(P<0.05),inula flowers and ground rice group differences without statistical significance.? Apoptosis effect research After building animal model group rats AM apoptosis percentage were significantly higher than the control group,28d and building more apparent,when compared with the control group with significant difference(P<0.01);7d,28d all give AM apoptosis percentage of a rat medicine group were significantly lower than the model group,compared with the model group had significant difference(P<0.01);Compared with when making mould 7d,28d AM apoptosis percentage of each drug group were significantly increased.? Angiogenesis effect researchAfter building animal model group rats,MIP-la/MCP-1 expression levels were significantly higher than that of control group,compared with the control group with significant difference(P<0.01).7d,28d each dosage group of rats,MIP-1?/MCP-1 expression levels were significantly lower than the model group,compared with the model group had significant difference(P<0.01).7d,28d fructus viticis,inula flower group,MIP-la/MCP-1 expression levels were significantly higher than in model group,compared with ground rice group,with significant difference(P<0.01).Can be seen from the above data,in the process of the pathogenesis of idiopathic pulmonary fibrosis,fructus viticis,inula flower can inhibit the expression of chemokines in order to improve the development of the disease.Rats after building,model group bFGF expression quantity increases,compared with the control group with significant difference(P<0.01);7d,28d each dosage group bFGF expression in rat serum were significantly lower than the model group,compared with model group have significant difference(P<0.01);7d,28d fructus viticis group bFGF expression quantity,inula flower group were significantly higher than in m group,compared with ground rice group,with significant difference(P<0.01).Rats after building model group PF4 expression levels were significantly lower than the control group,compared with the control group with significant difference(P<0.01);7d,28d each dosage group PF4 expression content in serum of rats were significantly higher than that of model group,compared with model group have significant difference(P<0.01).7d,28d fructus viticis PF4 expression quantity group,inula flower group were significantly lower than in m group,compared with ground rice group,with significant difference(P<0.01).Rats after building,compared with control group,model group of MVD in the lung tissue expression level in 7d,28d were significantly higher,has obvious difference(P<0.01).Compared with model group,fructus viticis,inula flower group and DXM group of lung tissue MVD expression significantly decreased when the 7d,difference was statistically significant(P<0.05);28d fructus viticis,inula flower group and DXM group of lung tissue MVD expression level are lower,but inula group and DXM group decreases more significantly,compared with the model group with significant difference(P<0.05).Rats after building,pulmonary arteriolar wall thickness in the 7d,28d were significantly thickened,when compared with control group,with significant difference(P<0.01),and 28d is the thickness of the 7d increase more obvious.For 7d,compared with model group,the thickness increase dosage group were reduced,fructus viticis,inula flower group of pulmonary arteriolar wall thickening has obvious improvement effect,difference was statistically significant(P<0.05);To 28d,compared with model group,fructus viticis,inula flower group,DXM group were significantly inhibit the IPF degree of rat pulmonary arteriolar wall thickening(P<0.01).ConclusionThrough the literature review,it is concluded that the combination of the lifting and the lowering of the body of the lung can improve the symptoms of pulmonary fibrosis and delay the progression of the disease.In the course of idiopathic pulmonary fibrosis,inhibition of apoptosis and angiogenesis is critical to controlling the disease.Applied experiment research,in the role of histopathologic study of each group rats,found in the process of pulmonary fibrosis hydroxyproline expression levels,increase collagen synthesis and deposition.Fructus viticis period,inula spent 7d alveolitis and pulmonary fibrosis are available in different degrees during the period of 28d inhibit lung tissue hydroxyproline content expression level,to intervene in the pulmonary fibrosis,but there are differences between the two targets,including floating drug fructus viticis stage,the main role in alveolitis and subsidence medicine inula main role in fibrosis stage;Found in the study of the role of cell apoptosis,fructus viticis,inula flower can inhibit apoptosis,and 7d inula flowers fructus viticis apoptosis inhibition effect is more obvious;28days,inula flower inhibiting effect more apparent than fructus viticis AM apoptosis,show that floating medicines and drugs may affect pulmonary fibrosis with timeliness;Found in the research on the role of angiogenesis,fructus viticis,inula flower and MIP-la can improve chemokines MCP-1 alpha expression level,by inhibiting the expression of chemokine gathered to suppress the secretion of inflammatory cells and suppressing alveolitis development.Fructus viticis,inula flower in the stage of alveolitis and pulmonary fibrosis stage all can inhibit bFGF,promote PF4 expression,thus inhibiting angiogenesis,and the inhibitory effect is more obvious in pulmonary fibrosis phase;At the same time can both reduce the microvascular density in different degrees,inhibit lung small artery wall thickness increases.Due to the different medicinal,a liter of a drop,role in stages of alveolitis and pulmonary fibrosis are different,the floating drug fructus viticis main role in the early stage of alveolitis,medicine and subsidence inula take main effect in the last stage of pulmonary fibrosis.The study found that the effects of the rise and fall of the vessel on the new blood vessels were confirmed by the results of the study.The effect of the drug is different,which can lead to different blood vessels.In addition,found AM apoptosis core role in angiogenesis,apoptosis starts to apoptosis of alveolar macrophages can make gathered a large number of secretion of chemokines,cytokines lead to disorder,resulting in angiogenesis.The pathogenesis of pulmonary fibrosis are mainly the prior period,therefore,excessive apoptosis and angiogenesis in the whole course of disease,excessive lead to inflammatory cells gathered themselves together,and developed into the pulmonary fibrosis.The use of lifting and lifting can delay the process of fibrosis by inhibiting the release of apoptosis and angiogenic factors.Improved pulmonary fibrosis can be used to improve the clinical medicine.
