Study On The Chemical Constituents Of Chinese Traditional Medicine Fructus Viticis

Objective:Fructus Viticis is contained in the 2015 edition of the Chinese Pharmacopoeia,which is a traditional Chinese medicine.It is a dry mature fruit of Vertexaceae,Vitex rotundifolia L.or Vitex trifolia L.It is widely used in China for medicinal purposes.In this study,we studied the chemical constituents of Fructus viticis and found that it has anti-hepatocarcinal active ingredients in Fructus viticis.It has screened out monomeric compounds that significant inhibition on the self-renewal of liver cancer stem-like cells,thereby providing an idea for finding an effective component against liver cancer from natural medicines.Methods:Separation and purification of fractions by modern chromatographic separation techniques such as silica gel column,sephadex LH-20 and high performance liquid chromatography?HPLC?,and nuclear magnetic resonance spectroscopy(¹H-NMR?¹³C-NMR?¹H-¹HCOSY?HMQC?HMBC)and high resolution mass spectrometry?HR-ESI-MS?technique was used to analyze the structure of the isolated compounds.After obtaining the monomeric compound,the compound obtained from the extract of the ethyl acetate extract of Fructus viticis was screened for anti-hepatocarcinoma activity by tumor sphere formation method,and the different concentrations?0,5,10,20?g/mL?of Fructus viticis were used.The active component of the ester moiety was used to treat stem cells of human liver cancer MHCC97-H cell line,and a monomeric compound having a significant inhibition of self-renewal and proliferation of liver cancer stem-like cells was selected.Results:Fifteen compounds were isolated and purified from Fructus Viticis,and one of the new secoiridoid compounds?Compound 1?.Nine monomeric compounds were screened for anti-hepatocarcinoma activity,including seven active components against hepatocellular carcinoma,three compounds of which have significant effects?Compound 6,7 and9?.Compound 1*can inhibit the self-renewal of liver cancer stem-like cells?LCSLCs?from human hepatocellular carcinoma MHCC97-H cell line.Conclusions:15 compounds were isolated,purified and identified from the ethyl acetate fraction of Fructus Viticis,including 9 lignan compounds,compound 1*is a new secoiridoid compound.The 9compounds were screened for activity,and there were 7 active ingredients against hepatocellular carcinoma.The inhibition of compound 6,7 and 9exhibited strongest activity.
