Purpose:The research purpose of perimenopausal depression is a female one of the highest incidence rates of disease,the pathogenesis and the hypothalamic pituitary adrenal axis(HPA axis)and the hypothalamic pituitary ovarian axis(HPO axis)endocrine hormone disorder related to this topic by bilateral ovariectomy(Ovariectomy,OVX)with chronic mild not predicted stress(CUMS)composite model of two steps to establish perimenopausal depression,under the guidance of Chinese medicine theory,study the effect of electroacupuncture on estradiol perimenopausal depression model rats serum(E2)and luteinizing hormone(LH),follicle stimulating hormone(FSH),gonadotropin-releasing hormone(Gn RH),procorticotropin releasing hormone(CRH),adrenocorticotropic hormone(ACTH)and corticosterone(CORT),?-endorphin(?-EP)effects of hormones,study of electroacupuncture therapy on perimenopausal suppression.Behavior of depression rats and the correlation between HPO axis and HPA axis,to explore the mechanism of electroacupuncture treatment of depression in perimenopausal period,to provide experimental basis for the theory and clinical application of traditional Chinese medicine in prevention and treatment of perimenopausal depression.Material and method:60 SPF grade SD female rats were randomly divided into control(control)group,model(model)group,sham-operation(sham)group,clomipramine(clomipramine)groupand the electroacupuncture(EA)group with 12 rats in each group.Establishment of a rat model of depression in the perimenopausal period by the combination of bilateral ovarian resection and chronic non predictable stimulation.Electroacupuncture group acupuncture "Shenshu"(BL23),"Sanyinjiao"(SP6),"Baihui"(GV20),1 times daily for 28 days;the clomipramine group with clomipramine hydrochloride intragastric administration,1 times daily for 28 consecutive days.After the treatment,observe the general condition of rats,changes of estrous cycle,sucrose preference,tail suspension test,forced swimming test results,using enzyme-linked immunosorbent assay(Elisa),the serum estradiol(E2)and luteinizing hormone(LH),follicle stimulating hormone(FSH),gonadotropin release hormone(Gn RH)and corticotropin releasing hormone(CRH),adrenocorticotropic hormone(ACTH)and corticosterone(CORT),?-endorphin(?-EP)were detected and analyzedResults: 1.Behavior changes: after modeling the other group than the control group,sugar consumption rate decreased(P < 0.05)and sham operation group after treatment,sugar consumption is lower than the model group increased(P < 0.05),electroacupuncture group and clomipramine group than in the model group increased sugar consumption rate(P < 0.05);model the other group than the control group,tail suspension test and the forced swimming test in rats with real time increased(P < 0.05),but after treatment,sham operation group,electroacupuncture group and clomipramine group the forced swimming test and tail suspension test in the immobility time of the rats was lower than that of model group(P < 0.05).2.Experimental index: after treatment,compared with model group,electroacupuncture group,clomipramine group and sham operation group,E2??-EP was higher than that in the model group(P < 0.05);electroacupuncture group,clomipramine group and sham operation group of LH? FSH?Gn RH ?CRH?ACTH?CORT were lower than the model group(P < 0.05).Conclusion: 1.Electroacupuncture therapy can adjust perimenopausal depression model of rat hypothalamic pituitary ovarian axis(HPO axis)and the hypothalamic pituitary adrenal axis(HPA axis),improve the behavior changes of perimenopausal depression in rats 2.Perimenopausal depression modelof hypothalamic pituitary ovarian axis(HPO axis)and the hypothalamic pituitary adrenal axis(HPA axis)inherent correlation,electroacupuncture endorphin content to achieve the treatment of perimenopausal depression model rats by improving.
