chapter,the effects of guiluerxianjiao were evaluated on dexamethasone induced osteogenic cell injury.The research method is as follows,the serum in the preparation process of SD rats were randomly divided into four groups:control group,ossification in three alcohol group,guiluerxianjiao low,high dose group,20 rats in each group.The blank group were not injected,guiluerxianjiao low dose and high dose of 0.05 g ml-1 0.1 g·ml-1,the positive control group was administered with ossification in three alcohol 0.1 μg kg-1.MC3T3-E1 osteoblasts were grouped into control group,model group(10-8 dexamethasone mol·L-1),ossification in three alcohol group,guiluerxianjiao serum containing low dose group,guiluerxianjiao Medicated Blood high dose group.Used to detect the MTT cell viability,cell supernatant was detected SOD and MDA activity in the supernatant was IL-1,IL-6,TNF-,inflammatory factor beta alpha level;using Western blot method to detect Nrf-2/HO-1/NF-protein expression level of B pathway.The results of the study showed that guiluerxianjiao serum containing low and high dose with ossification in three alcohol containing serum could significantly increase the osteogenic activity;at the same time,guiluerxianjiao serum containing low and high dose significantly increased osteoblast cell supernatant SOD content,reduce the content of MDA decreased;osteoblasts were inflammatory cytokines IL-6,TNF-the level of Western alpha and IL-1 beta;blot test showed that guiluerxianjiao serum containing low and high dose with ossification in three alcohol containing serum group were significantly increased osteoblast Nrf-2,HO-1 protein expression,decreased expression of p-I kappa B,P-NF-kappa B protein.Therefore,guiluerxianjiao bone cell injury induced by dexamethasone significantly improved,and the mechanism might be related with Nrf-2/HO-1/NF-kappa B pathway.In the second chapter,the effects of guiluerxianjiao were evaluated on osteoporosis rats induced by dexamethasone.The research methods are as follows:the experimental animal SD rats were randomly divided into five groups,except for the blank control group,the model group and the treatment group were given 5 mg·kg-1 dexamethasone intramuscular injection,once a week,continuous injection for 12 weeks to establish an animal model of osteoporosis rats.From the beginning of the sixth week,the drug group were lavaged with ossification in three alcohol(0.1 g·kg-1)and guiluerxianjiao low dose(0.5 g·kg-1),high dose(1 g·kg-1),the corresponding volume of normal saline with the blank and model group,for 6 consecutive weeks.After the end of the experiment,measurement of bone mineral density of rat fifth lumbar spine and left femur(BMD),serum phosphate,calcitonin,estradiol,determination of serum SOD,MDA assay,serum inflammatory factor IL-6,TNF-alpha,IL-1 beta level determination;pathological femoral structure changes were observed with HE staining,Western blot Turtle experiment deer antlers glue on rat synovial membrane protein Nrf-2,HO-1,p-IkBa,P-NF-kappa B.The experimental results show that guiluerxianjiao can significantly increase the bone density of femur in rats;guiluerxianjiao low and high dose serum bone alkaline phosphatase levels decreased significantly,calcitonin increased significantly,estradiol increased significantly.Guiluerxianjiao serum containing low and high dose with ossification in three alcohol containing serum could significantly increase the content of serum SOD and decrease the content of MDA,and can significantly reduce serum IL-6,TNF-alpha,IL-1 beta level.HE results showed that guiluerxianjiao serum containing low and high dose with ossification in three alcohol containing serum group,the formation of new bone trabecular and trabecular structure compared with the model group;Western blot showed guiluerxianjiao serum containing low and high dose with ossification in three alcohol group can significantly increase the expression of serum HO-1 protein,rat synovial Nrf-2,reduce the expression of p-I kappa B,P-NF-kappa B protein.Therefore,guiluerxianjiao can induce significant improvement in dexamethasone induced osteoporosis rats,and its mechanism may be related to Nrf-2/HO-1/NF-kappa B pathway.In summary,the study found that guiluerxianjiao on dexamethasone induced damage of osteoblasts,and dexamethasone induced osteoporosis rats had obvious improvement,the mechanism of its-anti-inflammatory and antioxidant activity related.A further study found guiluerxianjiao of osteoporosis may be related with Nrf-2/HO-1/NF-kappa B pathway.These results will lay a theoretical foundation for the prevention and treatment of guiluerxianjiao dexamethasone induced osteoporosis rats. |
