Non-alcoholic fatty liver disease(NAFLD)is one of the most common chronic liver diseases in the world.It is associated with clinical conditions such as obesity,insulin resistance(IR)and type 2 diabetes.Liver changes range from simple liver steatosis to non-alcoholic steatohepatitis(NASH),cirrhosis and hepatocellular carcinoma.The pathogenesis of NAFLD has been confirmed,such as metabolic disorders,lipid accumulation,insulin resistance and inflammation,but its underlying mechanisms,including those that drive disease progression,are not fully understood.Blood-letting therapy(hereinafter referred to as"pricking blood therapy")is one of the traditional characteristics of traditional Chinese medicine,its origin can be traced back to the Neolithic Age,the ancient name of "Prickling”.The research group has found that the pricking blood therapy has a significant therapeutic effect on non-alcoholic fatty liver in many years of clinical practice.To explore the efficacy and possible mechanism of blood-letting intervention on non-alcoholic fatty liver disease,this study was established with a high-fat diet feeding method.Non-alcoholic fatty liver rat model,using enzyme-linked immunosorbent assay for insulin resistance-related protein content as an observation index.By observing the changes in the serum levels of LP,ADP,FFA and CREP in the rats of the experimental groups,the influence of blood-letting intervention on non-alcoholic fatty liver rat model of insulin resistance was evaluated,and the "Fenglong"and "Taichong" were explored."The possible mechanism of action of blood-letting intervention therapy on non-alcoholic fatty liver disease model rats.This paper is divided into two parts:literature research and experimental research.The literature research includes three reviews,reviewl,the recognition of non-alcoholic fatty liver in the motherland medicine;review 2,the treatment progress of non-alcoholic fatty liver disease with pricking blood therapy;review 3,the modern research progress of non-alcoholic fatty liver.In the experimental study,a non-alcoholic fatty liver model was established by high-fat diet feeding therapy,and the effect of blood-letting intervention on non-alcoholic fatty liver model rats was evaluated.Objective:To observe the changes of LP,ADP,FFA,and CREP in the serum of experimental rats,and to evaluate the effects of blood-letting intervention on serum LP,ADP,FFA,and CREP of nonalcoholic fatty liver model rats.To explore the possible mechanism of the effect of "Fenglong" and "Taichong" blood-letting intervention therapy on non-alcoholic fatty liver disease model rats.Methods:The preparation formula of high-fat feed such as Chen Shiqing was used to feed high-fat mixed feed consisting of 10%lard,2%cholesterol and 88%normal feed.Fatty liver rat models were established by a high fat diet for 8 weeks.40 clean male SD rats were randomly divided into normal group(n=10),model group(n= 10),easy Shanfu group(n=10),and pricking blood intervention group(n= 10).Results:After 12 weeks of the experiment,each group of rats was weighed and compared.Compared with the normal group,the model control group,the puncture blood intervention group,and the Yishanfu group had lower body weight,and the difference was statistically significant(P<0.05).Compared with the normal group,the liver index of each group was significantly higher in the model group(P<0.05).The bloodletting intervention group and Yishanfu group were slightly higher,but the difference was not statistically significant(P>0.05);Compared with Yishanfu group,there was no significant difference in hepatic index between the bloodletting intervention group(P>0.05).After 12 weeks of experiment,serum leptin(LP),free fatty acids(FFA),and cAMP response element binding protein(CREP)levels were significantly higher in the model group than in the normal group.Adiponectin(ADP)content was significantly decreased in the model group;In group comparison,the levels of LP,FFA,and CREP decreased significantly in the drug group and the LPS intervention group,and the serum ADP content increased significantly.Compared with the drug group,the LPS,FFA,CREP content in the LPS intervention group was slightly higher,and the serum ADP content was slightly lower.Conclusions:This study used "Fenglong" and "Taichong" to treat non-alcoholic fatty liver model rats with blood-letting intervention on the treatment of non-alcoholic fatty liver disease.The mechanism may be through the improvement of NAFLD.The level of ADP in the liver of rats reduced the content of LP,FFA,and CREP,and promoted the body’s own ability to improve IR,which played a role in reducing the deposition of hepatic lipids and preventing the progression of fatty liver.
