The paper is formed by two parts:literature review and experimental study.First section:literature reviewThere are two literature reviews.The first is focused on the application and study about animal model of spinal cord injury(SCI)for several decades in the world.There are five section in the article:the establishment of the model,the selection of experimental animals,the location of injury,Classification of animal models of spinal cord injury and summary of the animal model of spinal cord injury.The second mainly states the knowledge about spinal cord injury from the point of modern medicine and traditional Chinese medicine,at the same time,introduces the achievements of our research group on Upper cervical spinal cord injury.Second section:animal experimentObjective:To explore the effect and underlying mechanism of early decompression(DE)joint Governor Vessel(GV)electro-acupuncture(EA)and delay decompression(DE)joint Governor Vessel(GV)electro-acupuncture(EA)on rats with acute severe upper cervical spinal cord compression injury.Methods:the rats were randomly divided into seven groups:sham,12h SCI,12h GV EA(SCI+EA),12h methylprednisolone(MP)(SCI + MP),48h SCI,48h GV EA(SCI + EA)and 48h methylprednisolone(MP)(SCI + MP).Creating the models of acute severe upper cervical spinal cord compression injury by forcing a balloon catheter put in atlas pillow clearance.Compressing for 12h and 48h to simulate the early and delay decompression,treatment means contained GV EA and MP,sacrificing all the rats after GV EA treatment for 14d.(1).BBB score to evaluate the change of each group hind limbs motor function.(2)The ELISA method to assess the change of the content of platelet activating factor(PAF)in injured spinal cord tissue,(3).Western blot method to assess the Caspase9 expression for each group.Results:(1)BBB score:the sham groups were 21±0.00 points all the time at the 5 time points.In the 3 groups of compressing for 12h,there was no statistical difference(P>0.05)between the GV EA group and the MP group at all the time points,compared with the SCI group,the GV EA group and the MP group had a higher score(P<0.05)at 24h,3d,7d time points,there was no statistical difference(P>0.05)between the 3 groups at 14d time point.In the 3 groups of compressing for 48h,there was no statistical difference(P>0.05)between the GV EA group and the MP group at all the time points,compared with the SCI group,the GV EA group and the MP group had a higher score(P<0.05)at all the time points(2)The content of PAF,The change between the groups compressed for 12h and the groups compressed for 48h was accordant,The PAF contents of GV EA group was lower than that of the MP and SCI groups.The PAF contents of the MP groups were lower than that of the SCI groups.(3).The caspase-9 expression:The change between the groups compressed for 12h and the groups compressed for 48h was accordant,The caspase-9 expression of GV EA group was lower than that of the MP and SCI groups.The caspase-9 expression of the MP groups were lower than that of the SCI groups.Conclusion:the GV EA had proven effective for SCI,the delayed decompression joint GV EA had a more effective treatment than decompression only,the early decompression joint GV EA can speed the recovery of motor function than decompression only,but whether has a better final effect needs to be proved further.
