Objective:Using acute spinal cord injury model in adult rats,to observe the effect of electro-acupuncture on expression of Caspase-3 following acute spinal cord injury which is the neuronal apoptosis-related gene and the impact of changes in the law,further to reveal the mechanism of electro-acupuncture about acute spinal cord injury in adult rats.Method:Healthy female Wistar adult rats(clean grade),body weight:220±10g,A total of 200 rats(100 for paraffin section immunohistochemistry,100 for FQ-PCR,20 for electron microscopy)randomly divided into 4 groups,each group of 11:electric-acupuncture group,after injuried,give electric-acupuncture treatment;drug group:after injuried,give Caspase-3 inhibitor treatment;model control group,after injuried,give no treatment;sham-operation group,only exposed spinal cord,no injuried;each group is further divided into 6h,1d,3d,7d,14d time groups,each time group have 5 rats used to immunohistochemistry examination.Before executed,need to assess rats motor function by the method refers to the new standards of Basso,Beattie and Bresnahan(BBB)scale.To observe the morphological in spinal cord tissues by HE staining.and the expressions of Caspase-3 by immunohistochemical method.Process data by the SPSS 13.0 statistical analysis.Results:1.The effect of electro-acupuncture on ethology following acute spinal cord injury in adult rats:using the BBB score,there are eight score higher(72.73%),two score lower (18.18%),one died(9.09%)after electro-acupuncture;but there are three score higher (33.33%),six died(66.67%)in the drug group.The effect of electro-acupuncture on motor function is better than the drug group,but the risk of death is below then the drug group.2.The effect of electro-acupuncture on pathomorphology following acute spinal cord injury in adult rats:electro-acupuncture group and drug group compared with model group significantly alleviated the spinal cord organizes pathological change at various time.In the 6h electro-acupuncture group:neurons in better shape,central Nissl bodies disappeared,and marginate together,this phenomenon appeared in early ischemic after injuried.In the 6h drug group:neurons become smaller space and light edema.In the 1d electro-acupuncture group:Nissl bodies marginate together,neuron morphology better.In the 1d drug groups:neurons in better shape,central Nissl bodies disappeared,and marginate together.In the 3d electro-acupuncture group:central Nissl bodies disappeared and marginate together.In the 3d drug groups:the number of neurons increased,but glial cells decreased.In the 7d electro-acupuncture group:dead neuronal and glial cell decreased,central Nissl bodies disappeared and marginate together.In the 7d drug groups:neurons repaired,glial cells increased.In the 14d electro-acupuncture group:Nissl bodies gathered around the nuclear.In the 14d drug groups:less neuronal death,shape well,and the number of glial cells reduced.3.The effect of electro-acupuncture on expressions of Caspase-3:shown that in the same point in time,different processing factor reflect the differences between the groups as follows:in the sham-operated group positive cells expressed.In the model group,6h after the injury began to express,express higher in 3d,declined in 7d,14d.In 3d,the expression of the drug group is less than the model group,the model group was higher than sham-operated group(P<0.05);in the 14d,the expression of Caspase-3 in the electro-acupuncture group is less than the model group(P<0.05).Conclusion:1.Electro-acupuncture can promote hindlimb motor function recovery following ASCI in adult rats.2.Electro-acupuncture can reduce the damage of pathomorphology following ASCI in adult rats.3.Through inhibit the expression of Caspase-3,Electro-acupuncture can protect the function of spinal cord following ASCI in adult rats.
