Chinese medicine is an important part of Chinese civilization.It is the medical treasure that our Chinese nation has been struggling with illness for thousands years and the practice with theoretical experience was combining,accumulating and developing.In recent years,with the improvement of people's living standard and the transformation of health and wellness concepts,traditional Chinese medicine has entered people's field of vision and has been highly valued by the entire society.The combination of Chinese medicine and modern science and technology can make the unique charm and value of Chinese medicine be excavated.This paper established a method: macroporous absorption resin(MAR)was combined with prepared high performance liquid(PHPLC),optimized the extraction and choiced the purification of the effective ingredients of Radix Puerariae,Fructus Sophorae and Clove Fruit,combinating with high performance liquid(HPLC),liquid mass transfer application(HPLC-MS),ultraviolet spectrum(UV)analysis and structure identification.In this paper,we can provide help for the next step of large-scale purification and pharmacological research of pueraria and other traditional Chinese medicine.1.Using the macroporous adsorption resin and preparative high performance liquid chromatography to separation Radix Puerariae.The optimal extraction process of the components in Radix Puerariae was explored:60% of the methanol ultrasonic extraction was 15 times,and the extraction time was 1.5h,and the extraction time was 2 times.Through adsorption and analytical experiments,the physical and chemical properties of 6 kinds of macroporous adsorption resins were compared,and the suitable resin was selected for the initial separation of the components in the puerariae and the target components.Then type in the preparative high performance liquid chromatography extract of treated by resin purification,choose suitable for puerarin flavonoids PHPLC separation condition is on sample amount: 1.0 mL(20 mg),detection wavelength: 250 nm,mobile phase: acetonitrile-water(were 20:80,V/V)as mobile phase,such as degree of elution velocity: 30 mL/min,get a 3'-hydroxy puerarin,puerarin,3'-methoxy puerarin,puerarin wood indican,daidzein is tasted,the purity of 98.38%,99.65%,98.77%,96.56%,97.71%,and at the same time through ultraviolet spectrum,liquid mass and nuclear magnetic resonance spectrum of five kinds of material composition were determined.2.Macroporous adsorption resin and PHPLC was used to separate the components of fructus sophorae.The optimal extraction process of the components in the locust was explored: 90%ethanol was extracted from solvent reflux,and the liquid ratio was 1:10.The extraction time was 1 h,and the extraction time was 3 times.Through adsorption and analytical experiments,the physical and chemical properties of 7 kinds of macroporous adsorption resins were compared,and the adsorption capacity of the resin was preliminarily explored,and the initial separation of the target components in the locust Angle was selected.Type coupled with the preparative high performance liquid chromatography to separation of resin isolate points twice,get the genistein-7,4'-di-O-?-D-glucoside,genistein-7-O-?-Dglucopyranos--de-4'-O-[(?-L-rhamnopyransoyl)-(1-2)-?-D-glucopyranosede],kaempferol-3-O-?-L-ramnopyranosyl-(1-6)-?-D-glucopyranoside.genistein-4'-?-L-rhamnopyransoyl-(1-2)-?-D-glu--copyranoside,at the same time can be measured by high performance liquid chromatography purity of 96.7%,97.2%,97.8%,97.2%,in combination with liquid mass,ultraviolet and nuclear magnetic resonance spectrum of the four kinds of material composition were determined.3.Extraction process and analysis of phenol derivatives in Clove Fruit.Groping for creating the optimum extraction technology of Clove Fruit of composition: 70% ethanol as extraction solvent,the reflux extraction,and solid-liquid ratio of 1:10,each time for 1 h,extracting 3 times,ethyl acetate extract twice,to get crude extractings from Clove Fruit.Then,the preparative high performance liquid chromatography conditions was explored.The extracts of the Clove Fruit were separated,and eugenol and mother eugenol were obtained.The purity was 99.3% and 96.0%,and the composition was determined by UV and HPLC-MS.
