ObjectTo investigate the effects of different length of aerobic exercise on IL-4,IL-10 secretion and the phosphorylation of STAT6 in myocardial tissue of rats after myocardial infarction(MI),and the mechanism of STAT6 in inflammatory response.MethodsForty male Sprague-Dawley rats(8-week-old)were randomly divided into 3 groups,sham operation group(Sham),myocardial infarction group(MI),and MI plus exercise for 8 weeks(ME).The MI model was established by left anterior descending coronary artery(LAD)ligation,and the exercise mode was intermittent aerobic exercise.Exercise program included 1 week of adaptive exercise,then each day included 10 m/min warm up for 10 min,20 m/min exercise for 8min,and then 15 m/min exercise for 2 min.Intensity alternating 4 cycles,and finally 20 m/min exercise for 8min,10 m/min after 2 min exercise.It was training once a day,5 d/w,training for 8 week.The cardiac wall thickness and ejection function(EF)were measured by two-dimensional echocardiography.The levels of IL-4 and IL-10 in myocardium of rats were measured by ELISA.The expression of p-STAT6 and t-STAT6 in myocardium were detected by Western blot.Results(1)The ventricular wall of the MI group rat was got thinner,and the EF was significantly lower than that of Sham group(P?0.01).The ventricular wall thickness,ejection function was increased by 8 weeks of aerobic exercise,end-diastolic left ventricular posterior wall thickness(LVPWd)and end-diastolic ventricular septal of ME group rats was significantly higher than those of MI group's(P?0.05),EF of ME group was significantly higher than that of MI group(P?0.05).(2)The heart weight/body weight ratio(HW/BW)of MI group was significantly lower than that of Sham group(P?0.05),and the HW/BW of ME group were significantly increased than MI group(P<0.05).The lung weight/body weight ratio(LW/BW)of ME group was significantly lower than MI group(P? 0.01).(3)The level of IL-4 in myocardial tissue of ME group was significantly higher than that of MI group(P:?0.05).The levels of IL-10 in myocardium of ME groups rats were significantly higher than those of MI group(P?0.05).(4)It was showed by Western blot that the ratio of p-STAT6/t-STAT6 of ME group were significantly higher than that of MI group(P?0.05).Conclusions(1)Aerobic exercise is beneficial to the improvement of myocardial systolic and diastolic function in MI rats.(2)Aerobic exercise could help to recover heart function post MI,reduce inflammatory response.(3)Aerobic exercise could increase the secretion of IL-4 and IL-10 in myocardial tissue of MI rats.(4)Aerobic exercise could contribute to the activation of STAT6 in the rat myocardial tissue,thereby inhibit inflammatory response.
