Cell viability detection is an important research method in life science,and it has important technical value and wide application prospects.With the rapid development of research on conquer of major disease and the urgency to promote technological progress in life science,new situation proposes high demand for promotion of detection methods and equipment performance.Cell viability detection based on impedance analysis has been widely studied and applied with the advantages of non-invasive,and automated continuous detection.At present,cell viability detection based on impedance analysis uses cell impedance amplitude under certain frequency point to analyze viability.However,this method is significantly affected by different interference,and the detection stability and accuracy are not high.In addition,the high-cost impedance analyzer is used to measure cell impedance during detection,and the impedance data is manually derived for further measurement of cell viability.Therefore,the cost of detection is high,and real-time performance is at lower level.Because of the shortcomings of cell viability detection,the on-line detection system is designed to improve cell viability detection performance and achieve the goal of low-cost,on-line detection.The main research contents are as follows:(1)On the basis of the relevant theory of cell viability detection based on impedance analysis and using COMSOL simulation technology,this paper proceeds from four factors to analyze the performance of cell viability detection based on impedance analysis.The four factors are measurement noise of cell impedance,distribution of cell attachment on the electrode surface,cell impedance frequency characteristics and the stability of detection.The optimization measures and treatment methods are proposed in this paper.(2)A new method for detecting cell viability with soft-sensing is proposed.We extract three characteristic quantities from cell impedance spectroscopy,there are impedance spectrum peak value,impedance spectrum area,and high-frequency impedance magnitude.The feasibility of this method is analyzed,and extracting method of instrumental variable is provided.Afterwards,we elaborate the complete process of establishing a cell soft-sensing model based on the LS-SVM algorithm.(3)The device setup,software and hardware design for detection system are developed.The low-cost AD5933 chip is used as the impedance measurement core,and the WiFi communication technology is used to establish a reliable communication connection between the cell impedance acquisition terminal and the cell viability analysis terminal.And the relevant module performance of the system is analyzed.(4)Taking the detection system constructed in this project as object,sufficient training data is acquired to establish a special model of cell viability soft measurement by experiment,and then we compare and analyze the overall performance of the system from the aspects of continuous detection stability and detection accuracy.Compared with the traditional method,the comparative experiment shows that the cell viability soft measurement based on impedance analysis can reduce the impact of interference on the detection process,and effectively improve the performance of detection.The detection system is designed on the basis of optimization results of the impact analysis.Owing to fusing soft-sensing method,it maintains high detection stability and accuracy with low cost.The detection system achieves the goal of scatheless,real-time and on-line detection,and it also improves the automation level of cell viability detection.
