Cucumber is a popular vegetable and widely cultivated in the plastic-film greenhouse in China during cold seasons.The soil and rhizosphere harbor a high diversity of microorganisms,which is thought to play important roles in promoting cucumber health and growth.To promote soil improvement and establish new disease contrstrategies,the soil and cucumber-rhizosphere soil from plastic-film greenhouses for cucumber cultivation in Wuhan,Macheng and Jingzhou,Hubei Province were collected.The 16 s rRNA amplicon sequencing technology was used to analyze the structure and function of soil and cucumber rhizosphere microbiomes.Results of this study are summarized as follows:1.Analysis of structure and function of soil microbiome in plastic-film greenhouse resulted in structure of soil microbiome displayed obviously regional differences.It suggested that Proteobacteria and Actinobacteria were the most dominant phyla,Bacillus and Sphingomonas were the most dominant genus.The soil microbiome involved in various essential processes,which mostly included amino acid metabolism,carbohydrate metabolism and cell growth and death.2.The structure and function of rhizosphere microbiome were analyzed,suggesting that Proteobacteria and Bacteroidetes were the most dominant phyla,and Bacillus and Rhizobium were the most dominant genus.The structure of cucumber rhizosphere microbiome in plastic-film greenhouse displayed obviously regional differences,and presuming involved in various essential processes,which mostly included cell growth and death,signal transduction and cell motility.3.The result of structural and functional differences between rhizosphere and bulk soil microbiome analysis,showing that the microbial community diversity of bulk soil was significantly higher than that of rhizosphere soil.The relative abundance of Proteobacteria and Bacteroidetes was significantly higher in the rhizosphere soil,but that of Acidobacteria,Actinobacteria and Chloroflexi was significantly lower.Although the community structure of rhizosphere and bulk soil microbiome is different,their function is more conserved in evolution.4.By the same sequencing technology,core taxa of cucumber rhizosphere microbiome were analyzed.Results showed that 24 bacterial genera were identified in the core cucumber rhizosphere microbiome.The core rhizosphere microbes mostly included Rhizobium,Ensifer,Streptomyces,Devosia,Lysinibacillus,Novosphingobium and Dyadobacter.
