Protozoa is the third largest biota in the soil microbiome.Although the research on the community structure of soil protozoa has gradually increased,the ecological functions of protozoa and its relationship with other biota remain to be explored.In this study,the amplicon sequencing technology in high-throughput sequencing was used to compare and analyze the microbial community in rhizosphere soil of two crops,and the correlation between protozoa community,bacterial community and environmental factors was explored.The response of functional biota to nitrogen was preliminarily analyzed.The results are as follows:1.Temporal and spatial characteristics of microbiome community structure and diversity in rhizosphere of two crops were describedIn the rhizosphere soil of the two crops,protozoa are the third largest group except bacteria and fungi,and the dominant groups are Litostomatea,Colpodea and Phyllopharyngea.The dominant groups of bacteria include Actinobacteria,Proteobacteria,Acidobacteria,Chloroflexi,Gemmatimonadete and Bacteroidetes.The diversity index of protozoa and bacterial communities varied with growing period and the difference in the rhizosphere of twocrops is great.2.The correlation among protozoan community,bacterial community and environmental factors in crop rhizosphere was analyzedIn sugarbeet rhizosphere,A-P,TN and NO3--N were the main environmental factors affecting protozoan community,and TN and A-P were the main environmental factors affecting bacterial community.In potato rhizosphere,A-P and TOC were the main environmental factors affecting protozoan community,and T,p H,TN,A-P and NO3--N were the main environmental factors affecting bacterial community.There was a high correlation between B group,P group,A group,R group of sugarbeet rhizosphere protozoa and Actinobacteria,Proteobacteria,Acidobacteria.There was a high correlation between B group,P group,R group of potato rhizosphere protozoa and Actinobacteria,Proteobacteria,Acidobacteria.3.For 27 protozoa and 47 functional bacteria with high correlation with nitrogen,the response of functional biota to ammonia nitrogen,nitrate nitrogen,total nitrogen and C/N ratio was analyzed.7 species of protozoa and 11 species of bacteria were directly related to nitrogen.There were 2 species of protozoa and 6 species of bacteria indirectly related to nitrogen.Among them,there were 5 species of protozoa and 8 species of bacteria with positive correlation.There were 6 species of protozoa and 10 species of bacteria with negative correlation.These micro organisms have different effects on the geochemical cycle of nitrogen by participating in the metabolic process of soil nitrogen.
