Optically induced dielectrophoresis is a novel micro-nano manipulation technology developed on the basis of dielectrophoresis.Compared with traditional dielectrophoresis,optical tweezers and magnetic tweezers,optically induced dielectrophoresis can be used for real-time virtual electrode design according to requirements,and the use is more flexible.Optically induced dielectrophoresis is a non-contact,non-invasive manipulation that can be applied to biomedical cell screening,targeted therapy,separation,transport,and capture of particles in other fields,and has a good application prospect.This paper first introduces in detail the theoretical basis,advantages and disadvantages of several micro-nano manipulations,and then introduces the research progress and theoretical analysis of optically induced dielectrophoresis.The principle of manipulation of optically induced dielectrophoresis was derived based on the working mechanism of dielectrophoresis.Next,the characteristics and applications of magnetic nanoparticles and the preparation process of magnetic target cells were detailed,which laid a foundation for the subsequent preparation of mixed cell solutions.Finally,the optically induced dielectrophoresis chip was designed and the experimental system was set up to realize the manipulation of yeast cells and magnetic target cells based on optically induced dielectrophoresis,and the magnetic target cells were screened from the mixed cell solution.In this experiment,magnetic target cells and yeast cells were manipulated and screened by optically induced dielectrophoresis.The manipulation of the yeast cells and the magnetic target cells is achieved by changing the electrical conductivity of the photoconductive layer by light to form a virtual electrode while applying an alternating current signal.Then the strip electrode was designed to successfully screen the magnetic target cells,and the feasibility of screening the magnetic target cells by optically induced dielectrophoresis was verified.Finally,based on this,the effects of different experimental parameters on manipulation and screening of cells were studied,and the optimal conditions for screening magnetic target cells were obtained.
