Perfluorinated sulfonates(PFOS)and perfluorinated sulfonic acids(PFOA)are widely used in the titanium,waterproof materials,cosmetics,automobile coatings,etc.because of their hydrophobic,oleophobic and dustproof.However people found that PFOS and PFOA had toxicity and potential ecological risk,so the European Uniona and the United States had issued the restrictions of PFOS and PFOA.Planaria Dugesia japonica which had the simple nervous system and strong regeneration ability was sensitive to toxins.We studied the change of the genes which were related to nervous system and the acticity of enzyme in planarians which were exposed to PFOS and PFOA.The damage of nervous system was detected when regenerated planarian were exposed to PFOS and PFOA.1 The change of genes which were related to nervous system caused by PFOS and PFOA in planarian.Planarians were exposed to PFOS and PFOA respectively.We used qPCR to detect the change of five genes which were related to nervous system.Results showed that the expression of FoxD decreased with the exposure of PFOS and PFOA.PFOA had obvous impact on FoxG which expressed in substance of trunk.PFOS had obvious influence on FoxG which expressd in brain.The expression of otxA and otxB were suppressed significantly by PFOS.The expression of nlg was activated by PFOS and PFOA,but the promoting effects of PFOS had apparent lag.2 The change of activity of enzyme caused by PFOS and PFOA in regenerated planarian.Regenerated planarians were exposed to different concentration of PFOS and PFOA,we used spectrophotometry to measure the activity of the enzyme in planarians on the 1st,3rd,5th,7th and 10thh day of regeneration.The results showed the acticities of glutathione peroxidase(GST-Px),glutathione s transferase(GST),peroxidase(POD)and acetylcholinesterase(AchE)varied with the concentration of drug and the days of regeneration.The content of malondiadehyde(MDA)also had corresponding change.As a result,PFOS and PFOA had biochemical toxicity in planarian.3 The change of nervous system caused by PFOS and PFOA in regenerated planarianRegenerated planarians were exposed to different concentration of PFOS and PFOA.On the 1st,3rd,5th,7th and 10thh day of regeneration,we used hydrochloric acid to kill the planarian,paraformaldehyde to fix the planarian and immunohistochemical experiments to locate the synapses of planarians.The results showed that the regeneration processes of nervous system were affected by PFOS and PFOA.PFOA had impact on ventral nerve cord,PFOS had influence on cerebral ganglion and the nerve in tail.With the concentration increasing,the damage aggravated.
