Carassius auratus is rich in protein,sugar,fat and other nutrients and the protein content is higher.The protein of carassius auratus is a kind of high quality protein which has a variety of amino acids and a lot of essential amino acids.The medicinal value of carassius auratus is very high.The carassius auratus meat can prevent and treat arteriosclerosis,hypertension and coronary heart disease.It also has the effect of lowering cholesterol.Bioactive peptides prepared by enzymatic hydrolysis of food protein raw materials are easy to be absorbed by human body.The bioactive peptides have many biological functions such as antifatigue,blood pressure lowering,antioxidant,antithrombus and so on.Therefore,it is of great significance for the development of health care products from carassius auratus.The main research contents and results are as follows:The content of the basic components of carassius auratus was determined.The content of protein in carassius auratus is 18.06%.The fat content is 2.32%.The total sugar content is 3.17%.The ash content is 1.55%and the moisture content is 74.89%.Crude protein was prepared by acid extraction,alkali extraction,salt extraction and water extraction.The extraction rate and ACE inhibition rate of crude protein were compared.The results showed that the extraction rate of the four methods was low and the ACE inhibition rate of crude protein was very low.So we can see that the ACE inhibitory activity of the crude protein from carassius auratus is weak.The optimal protease was selected by the degree of hydrolysis and peptide content.The optimal protease was used to optimize the enzymatic hydrolysis process by single factor and orthogonal experiment.The optimal conditions of the enzymatic hydrolysis process were pH 8,enzyme hydrolysis temperature 53?,enzyme concentration 4000U/g.Gel chromatography and the glucan gel electrophoresis were adoped,aiming at separating the peptides and analyzing molecular weight distribution of the peptides.After gel chromatography,two components P1 and P2 were collected by freeze drying.Two components P1 and P2 were analyzed by Tricine-SDS-PAGE electrophoresis.The results showed that the molecular weight of PI was larger and the molecular weight of P2 was mainly in 8491 Da,3300Da and 5335-4006Da.The ACE inhibition rate of P1 and P2 was determined.The results showed that the inhibition rate of P2 was 66.42%.Linear regression was carried out on the different concentrations of P2 and their corresponding ACE inhibition rate.The IC50 value of P2 was 0.407 mg/mL.The preparation of polypeptide oral liquid was studied.The results show that the fishy flavor and bitterness is the lowest adding 2%activated carbon,3%beta cyclodextrin and 0.05%citric acid.Preliminary select 0.06%pectin,0.03%yellow gum as stabilizers.Then add citric acid,honey,xylitol,pectin,yellow gum for the preparation of oral liquid and sensory scores and stability indicators as the indexs.The optimal formulation is lemon acid 0.3%,honey 5%,xylitol 8%,pectin0.05%,yellow gum 0.04%.Finally,the quality indexs of the oral liquid of carassius auratus polypeptides were determined.The results showed that all the indexes reached the national standard.
