Macleaya cordata(Willd.)R.Br.is widely distributed in nature,which is also a kind of Insecticidal herbs commonly used in agriculture and animal husbandry.It contains kinds of alkaloids which are toxic in the body.And according to the previous research of the plant,Macleaya cordata(Willd.)RBr.Also has good sterilization effect on the Oncomelania hupensis.Therefore,this article aims to study the extractions of alkaloids of poison effect on Oncomelania hupensis and provide modified methods suitable for industry extraction and separation to develop a kind of great potential applications of plant source Oncomelania hupensis drugs.In this paper,acid water method and ethanol method were used to extract the total alkaloid in the leaves and pod of pod,respectively.The extraction rate of acid water method was 4.96%,8.07%respectively,and the extraction rate of ethanol method was 3.59%and 5.76%respectively.Therefore,the extraction effect of acid water method was obviously better than that of ethanol method.The extraction rate of the two methods all showed that the content of total alkaloids in the pod of the pod was obviously higher than that of the total alkaloids in the leaves.The results showed that the LC50 of total alkaloid and total base of pod in 72h were 182.1 mg/L and 25.1mg/L respectively,indicating that the total alkaloid from the pod of the pod was stronger than that from the total alkaloids from the leaves.Therefore,it is more convenient and effective to extract the effective components from the fruits of pomelo by acid water.Then,the total alkaloids in the fruits of pomphomiya fruit were further separated and purified,and the mixture of phenanthridine alkaloid and bisulfate was obtained.A high performance liquid chromatography-mass spectrometry(HPLC-MS)technique was used to analyze the total alkaloid and phenanthridine alkaloids of the pod of bloomoine.It was determined that the main alkaloids of the total base of the total alkaloid contained SA,CHE,PRO and ALL(ALL),and a small amount of berberine(BER).Anthridine alkaloid mainly contains(SA)sanguinarine and chelerythrine(CHE).The results showed that after treatment of 48h,72h,96h and 120h when the drug concentration was 2.5mg/L,the fatality rate of Oncomelania was 68.3%,91.7%,98.3%and 100%.After the drug concentration was 7.5mg/L,the fatality rate of Oncomelania was 100%,and the Oncomelania snails were soaked at 48h and 72h when the concentration of drug concentration was 7.5mg/L.The LC50 is 2.89 mg/L and 1.29mg/L,respectively.Moreover,under the concentration of 1 mg/L,the snail effect of the alkaloids of the alkaloids of the polycline is close to the niclosamide ethanolamine salt(snlosamide),which shows that the snail killing effect of the alkaloid of the faricidal alkaloid is equivalent to the snail killing effect of the main chemical snail,niclosamide ethanolamine salt(spiramine),which is the main chemical snail.The damage to the surface structure of the head foot and liver of Oncomelania Oncomelania was studied by SEM.It was found that the drug has a great damage effect on the epidermal structure of the head foot and liver of Oncomelania hupensis,especially the damage to the skin of the head foot,which is because the head foot is more easily exposed to the drug solution than the liver.In The damage to the internal structure of the head foot and liver of Oncomelania Oncomelania was studied by transmission electron microscope.It was found that the effects of drugs on the muscle cells in the head foot of Oncomelania snails were not significant,but the damage effect on the cells in the liver of Oncomelania snails was very significant.Show that the target organ of Macleaya alkaloids poisoning Oncomelania Feiding majorinliver.By measuring various enzyme activity indices of the liver cells of Oncomelania oncomelania,the effects of the alkaloids on the liver biological metabolism of Oncomelania snails were studied.It was found that the activity of glutathione S-transferase(GST),carboxylesterase(CarE)and alkaline phosphatase(AKP/ALP)in the liver of Oncomelania snails were treated at low concentration with the increase of treatment concentration.There is a trend of rising,and then in the high concentration treatment time or fast or slow down,and as the processing time increases,all kinds of indicators are in the early stage of a trend of increase,and then reprocessing later or fast or slow decline.This shows that Oncomelania Oncomelania in the early stage of drug treatment and in the lower concentration of the body's various kinds of biological metabolism began to be strengthened to resist the poison of the drug,and the metabolism of all kinds of biological metabolism in the later period of the drug treatment and at the higher concentration of the body weakened rapidly or slowly,indicating that the Oncomelania snails have been greatly affected.Injury.The results above show that Macleaya cordata(Will.)R.Br.is a highly potential plant which has good sterilization effect on the Oncomelania hupensis and pretty development value.The phenanthridine alkaloids extracted from the pod of Macleaya cordata(Willd.)R-Br.have excellent sterilization effect on the Oncomelania hupensis and are expected to be developed into a new type ofplant source Oncomelania hupensis drug.In this paper,we study the sterilization effect on the Oncomelania hupensis and the mechanism of phenanthridine alkaloids of Macleaya cordata(Willd.)R.Br.,hoping to lay a solid foundation for the future development of plant source Oncomelania hupensis drug researchs.
