Amantadine is an antiviral drug for human flu.In livestock and poultry husbandry,long-term and excessive use of amantadine can cause poisoning,directly inducing virus mutations,giving livestock and poultry a strong resistance,which not only affects animal quality,but also harms human health.Therefore,it is of great significance to quickly detect amantadine in real time.Amantadine does not fluoresce and can not be measured directly by ordinary fluorescence spectrophotometry.The cyclodextrin-like macrocyclic host molecule has the structural characteristics of"internal hydrophobic and external hydrophilic",and can contain alloy adanylamine and a series of fluorescent molecules.In this study,inclusion complexes of fluorescent molecules and cyclodextrin host were used as fluorescent probes for the detection of amantadine by competitive encapsulation.Most amantadine veterinary drugs are prepared by dissolving in water and mixing with the raw meal.Therefore,the content of amantadine in different brands of amantadine hydrochloride tablets was determined in this study,which has certain practical significance for the rapid detection of amantadine.The main research content is as follows:The fluorescence spectrophotometry was used to study the interaction between berberine and HP-?-CD.The results showed that berberine and HP-?-CD formed a 1:1 inclusion complex,and the inclusion complexes of them could produce strong fluorescence emission.Amantadine can quench the fluorescence produced by the inclusion complex of berberine and HP-?-CD.The determination of the amantadine content in the tablet was achieved by establishing a linear relationship between the fluorescent quenching value of the inclusion complex and the concentration of amantadine.The detection limit was 0.3 mg·L-1,the recovery rate was between 92%and 101%,and the relative standard deviation was less than 1%.Indicating that this method had good sensitivity and selectivity.Taking the strong binding ability of amantadine and?-cyclodextrin as a reference,the NBD-Cl molecule was connected to adamantyl,and the fluorescent molecule NBD-adamantane was designed and synthesized.The spectral properties of the compounds were studied.The results showed that NBD-adamantane had high sensitivity,and the fluorescence intensity changed significantly under different organic solvents and temperature conditions.The fluorescence intensity was the strongest at pH 4.38,which provided a basis for the inclusion of amantadine and cyclodextrin.Fluorescence spectrophotometry was used to study the interaction between amantadine and?-cyclodextrin.The results show that amantadine and?-cyclodextrin can form a 1:1 inclusion complex.At the same time,the inclusion constant K=1.28×104 L·mol-1.the entropy change of thermodynamic parameters,the enthalpy change and Gibbs free energy change were measured.And inclusion complexes of NBD-adamantanamine and?-cyclodextrin could produce strong fluorescence emission.The interaction of the inclusion complex with amantadine was studied.The results showed that amantadine quenched the fluorescence of the inclusion complex.Based on this,a fluorescent probe method for the determination of amantadine was established.At the same time,the method was applied to the determination of amantadine in tablets and good measurement results were obtained.
