Silane coupling agent is a common organic reagent whose molecules contain functional groups which can connect both organic and inorganic materials including magnetic nanoparticles and small organic molecules.Silane coupling agents acts as intermediay materials,the alkyloxy group could be hydrolysis by water to form silanol groups,then forms silicon oxygen bond with hydroxyl group on the surface of inorganic materials such as magnetic nanoparticles.The other functional groups end such as amino group,carboxyl group and sulfhydryl group,could react with all kinds of organic compounds and connect with organic material.The disulfide bond is a dynamic chemical bond.Under the action of reductant or oxidoreductase,the disulfidebond can be and reduced to sulfhydryl group.By introducing a cleavable disulfide bond in the silane coupling agent,the connected materials could be quicked cleaved after the conncetion.Silane coupling agent is a common magnetic nanoparticles modifier,which can control the size of nanoparticles,enhance the dispersion of particles,and introduce reactive groups on the surface of magnetic nanoparticles.The first part of the paper,A silane coupling agent containing a disulfide linked terminal azido group was synthesized by chemical method.The coupling agent can be used to modify magnetic nanoparticles.The second part of this paper,we also synthesized small organic molecules containing mercapto group,azido group,amino group and carboxylic acid group based on Polyethlene glycol.The azido group introduced can be clicked with the compounds containing the alkynyl group.The amino and carboxyl groups introduced can connect different proteins,and the oxygen amino groups can be used to oxime with aldehydes and ketones.These small organic molecules can be used for specific labeling and conjugation of proteins.Several bifunctional PEG compounds have been synthesized through different synthesis strategies.
