Compounds containing imidazo[1,2-?]pyridine structure are a kind of nitrogen-containing heterocycles with important biological activity,which are commonly found in natural product molecules and synthetic drug molecules.Considering the unique structural properties,biological activity of imidazo[1,2-?]pyridine compounds and their wide application value and their good pharmaceutical market prospects(such as zolpidem used as a sedative agent and apyrazole used as a anxiolytic agent),the synthesis and derivatization of these compounds have attracted more and more attention among chemical community.This paper is mainly divided into three chapters.In the first chapter,we briefly review the research progress of imidazo[1,2-?]pyridine compounds around the world in recent years,including the brief introduction of the biological activity of the compounds and the progress of the related derivatization reactions.The important application of the imidazo[1,2-?]pyridine motifs is highlighted.In the second chapter,the regioselective C-3 methylsulfanylation of imidazo[1,2-?]pyridines through diiodine mediated,acetone-promoted,C–S bond construction with dimethyl sulfoxide as both the source of the methylsulfanyl moiety and the solvent is described.The protocol features mild operating conditions,broad substrate scope and good functional group compatibility.In the third chapter,an expeditious and straightforward approach for the synthesis of 4H-pyrido[1,2-?][1,3,5]triazin-4-ones starting from imidazo[1,2-?]pyridines and sodium azide is presented under aerobic oxidative conditions.The reaction proceeded well with the promotion of K2S2O8/KMn O4 with good functional group tolerance.[1,3,5]triazin-4-one molecule skeletons are a kind of important compounds with specific biological activities,which have a wide range of applications in the fields of organic synthesis,biology and medicine,and there arefew reports about existing synthetic methods.Therefore,Notable features of this methodology include the usage of readily available materials,a broad substrate scope and easy scalability.
