The flavonoids in ginkgo biloba have anti-oxidation,anti-atherosclerosis,and blood circulation and other anti-cardio-cerebral vascular effects.The flavonoids and saponins in gynostemma pentaphyllum have the functions of lowering blood sugar,lowering blood fat,lowering blood pressure,and regulating the immune system in both directions.Trimetazidine hydrochloride has the effect of maintaining the energy metabolism of the heart and nerve sensory organs in ischemia and hypoxia.When ginkgo biloba and gynostemma pentaphyllum are compatible,their effects against cardiovascular and cerebrovascular diseases can be mutually enhanced.In addition,gynostemma can make up for the deficiency of single ginkgo leaf preparations by regulating blood lipids,blood glucose,lipid metabolism and immune function.This study based on the compatibility of traditional chinese medicine ginkgo and gynostemma,join the chemical medicine trimetazidine hydrochloride,give full play to the synergy of TCM plus western medicine.The purpose of the study was to prepare a new type of anti-cardiovascular and cerebrovascular diseases injection with less side effects and good curative effect.The main content of this study were as follows.1.In the condition of ultrasonic oscillation,the ultrafiltration method was used to extract anti-cardiovascular active ingredients of Ginkgo biloba and Gynostemma pentaphyllum.The optimum conditions for the ultrafiltration extraction of active ingredients in ginkgo biloba and gynostemma pentaphyllum were as follows:with 80%ethanol concentration,the ratio of solid to liquid was 1:20,and the ultrasonic time was 10 min.2.The active ingredients of Ginkgo flavonoids,Gynostemma flavonoids,and Gynostemma saponin were further purified by AB-8macroporous adsorption resin.The optimum purification conditions for AB-8 macroporous adsorption resin were as follows:The concentration of total flavonoids in the sample solution was 1.25 mg/mL2.50 mg/m L,the total saponin was 0.43 mg/mL1.29 mg/mL,at this time the adsorption rate can more than 70%.10%ethanol was used as a decontamination before desorption,70%ethanol was used as eluent,and the maximum sample size was 450 mL.The purity of total flavonoids and total saponins from purification were 37.35%,12.75%,and the ratio of flavonoids and saponins was 2.93.3.The prescription of the compound double chinese medicine ginkgo gynostemma trimetizidine injection were determined.The injection prescription:Purity 2.68 mgmL-1(equivalent to 1.0 mgmL-1flavone,0.3412 mgmL-1saponin),trimetazidine hydrochloride 0.4mgmL-1,propylene glycol 0.08 mLmL-1,dehydrated alcohol ethanol 0.12mLmL-1,sodium hydrogen sulfite 0.2%gmL-1.4.The stability of the preparation of compound double traditional Chinese medicine ginkgo ginkgo was investigated.The results of the injection stability shows that the injection is unstable when placed at60?,the content of the active ingredient changes within 5%for 30 days at-10?and 25?,which was in line with the“2015 Chinese Pharmacopoeia Stability Guidelines”.
