Pleurotus tuber-rigium(Fr.)Sing is a kind of rare edible fungus and medicinal bacterium that can produce large germ nucleus and fructification.Its fruit bodies and sclerotia are rich in nutrients,containing volatile oils,amino acids,and polysaccharides,of which the most active ingredient is the polysaccharide in Pleurotus tuber-regium(Fr.)Sing.Because of its good taste and unique flavor,the fructification of the pleurotus tuber-regium is favored by the public.However,the sclerotium has a loose texture and poor taste,which leads to low pubilc acceptance and lower utilization than the fruiting body.Raising the utilization rate of the Sclerotium of the Pleurotus tuber-rigium(Fr.)Sing can not only achieve its high value,but also create higher economic and social benefits.In this paper,water-soluble polysaccharide were extracted from pleurotus tuber-regium(Fr.)Sing Sclerotium by water extraction and alcohol precipitation method.We evaluated the effect of conditions for preparing the polysaccharide of pleurotus tuber-regium(Fr.)Sing Sclerotium by the yield of polysaccharides,and optimized by response surface methodology.SGC-7901 of gastric cancer cells cultured in vitro to study the anti-gastric cancer activity of polysaccharide from water soluble pleurotus tuber-regium(Fr.)Sing Sclerotium.The main research contents and conclusions are as follows:1.In this paper,the polysaccharide from the core of pleurotus tuber-regium was extracted by water and alcohol precipitation method,and the chemical and physical properties of polysaccharide were detected by Savge and Dialysis.The results showed that the polysaccharide content of the polysaccharide was 73.9%.It contained two main components and their average relative molecular weight are 111092.9Da and28298.4Da,respectively.2.With the polysaccharide yield as the index,the extraction temperature,extraction time,and material-liquid ratio were selected to optimize the extraction conditions of the water-soluble polysaccharide from pleurotus tuber-regium(Fr.)Sing Sclerotium.The optimal extracting process parameters were as followed:material/liquid ratio of 1:29,temperature 98 ?,extraction time 5 h.3.The cell proliferation rate and early apoptosis of gastric cancer cell line SGC-7901 were used to study the in vitro anti-gastric activity of water-solublepolysaccharides from pleurotus tuber-regium.The effect of different concentrations of water soluble pleurotus tuber-regium polysaccharide solution on proliferation rate and early apoptosis of gastric cancer cells by using WST-1 cell proliferation assay kit and flow cytometry.The results showed that the water-soluble polysaccharide of pleurotus tuber-regium(Fr.)Sing Sclerotium could promote the early apoptosis of gastric cancer cell SGC-7901 when the concentration reached 20 ?g/mL,and it could inhibit the proliferation of SGC-7901 at 60 ?g/mL affter 24 h.And the effects are enhanced with increasing concentrations of polysaccharides.This means that water-soluble polysaccharide of pleurotus tuber-regium(Fr.)Sing Sclerotium has a certain anti-gastric activity in vitro.4.This experiment used the sclerotium of pleurotus tuber-regium(Fr.)Sing as the raw material,through the use of the crushing granulation and other technical means to develop a edible solid beverage of pleurotus tuber-regium(Fr.)Sing Sclerotium,and clarified formula of the solid drink: 47.6% powder of pleurotus tuber-regium(Fr.)Sing Sclerotium,47.6% maltodextrin and 4.5%konjac glucomannan.
