Korean pine is one of the main forest species in northeastern forest areas in our country,its wood,bark,and seeds have been exploited,however Korean pine scale as waste treatment after the removal of pine nuts,has caused a waste of resources.In this study,Korean pine scale polyphenol as raw material,the mechanism of hypoglycemic action of Korean pine scale polyphenol was studied,Korean pine scale was used develop Korean pine scale polyphenols Effervescent Granules,and measuring the antioxidant activity of the product.Provide a new way for the development and utilization of pine seed scales.In this study,extraction and purification from Korean pine scale to obtain polyphenols.The Caco-2 cell monolayer model was used to simulate the intestinal epithelial cells,and the effects of Korean pine scale polyphenols on glucose transport were explored.By Caco-2 cell monolayer model,using 5mM and 25mM glucose simulated fasting blood glucose levels before meals and postprandial blood glucose levels,respectively,explored different concentrations of pine polyphenols affect glucose transport at different times,and mRNA expressions of the glucose transporter protein 2(GLUT-2)and sodium-glucose co-transporter-1(SGLT-1)were also studied.The results showed that glucose transport has a significant inhibitory effect when 100?g/mL polyphenols incubated Caco-2 cell monolayer model for one hour,inhibition rate was(11.572 ± 0.399)%at low glucose levels;inhibition at high glucose levels was(15.968 ± 0.347)%;and Korean pine scale polyphenols significantly reduced GLUT-2 and SGLT-1 mRNA expressions.The results indicated that by inhibiting glucose transport and GLUT-2,SGLT-1 mRNA expression,Korean pine scale polyphenols could modulating post-prandial blood glucose in humans,and could be further developed into healthful products for preventing and treating diabetes.Sodium bicarbonate is wrapped with polyethylene glycol 6000(PEG6000)by 1.0:0.3,and mannitol,citric acid,and other accessories were added to Korean pine scale polyphenols to produce the effervescent granules.Response surface methodology was used to optimize the formulation of composite parameters.The results showed that optimal technical conditions were:the ratio of mannitol and dextrin was2.15:1,the sorbitol addition was 10g/100g,the ratio of citric acid and sodium bicarbonate was 1.44:1.Under optimal condition,the sensory evaluation score was the highest,up to 80.64.The total polyphenols content of Korean pine scale polyphenols effervescent granules was studied,and calculate the recovery rate.The antioxidant activities of Korean pine scale polyphenols effervescent granules were assayed through the capability of the reducing ability,the capability of scavenging hydroxyl radical and the capability of scavenging DPPH.Inhibition kinetics of a-glucosidase by Korean pine scale polyphenols effervescent granules was studied,to research its hypoglycemic activity.The results showed that the polyphenols content of the effervescent granules was 13.76mg/g,and it showed high antioxidant activity.It has a strong DPPH radical and hydroxyl radical scavenging activity,IC50 values were 14.89 and 52.29mg/mL,respectively.The product has a a-glucosidase activity inhibition,IC50 value was 68.72mg/mL.This Korean pine scale polyphenols effervescent granules has a high content of polyphenols,its a function food with anti-oxidation and hypoglycemic activity.
