Korean pine seed oil is a kind of natural plant oil which is rich in unsaturated fatty acid, such as linoleic acid and linolenic acid, and has the functions of various health cares, such as losing weight, decresing blood fat, anti-aging, and increasing immunity etc. However, Korean pine seed oil is unstable and vulnerable to environmental factors because its molecules contain lots of conjugated double bonds, which improves its oxidation; therefore the nutrition and physiologically active function of pine seed oil can not be unutilized efficiently, and its applications are restricted greatly. Microencapsulation can protect pine seed oil from easily affecting by the environmental factors, improve its stability and make it more efficient in its functions.Korean pine seed oil was selected as an objective of study, the β-cyclodextrin(β-CD), malt dextrin and Auricularia auricula polysaccharide was selected as wall material, molecular embedding method and emulsion polymerization method were used to prepare the pine seed oil microcapsules respectively. The microcapsules were evaluated by determining encapsulation efficiency and oil load. Two different method of making pine seed oil microcapsule were compared, and use response surface analysis to optimize each technology. The best process condition of pine seed oil microcapsules prepared by molecular embedding is ratio of core and wall material of 1:4.9, time 93 min, temperature 49.3℃ and stirring speed 609r/min, under the technological conditions encapsulation efficiency can reach 66.38%, average oil load is 2.85%. The optimum process condition of pine seed oil microcapsules prepared by emulsion polymerization is solid content 15.5%, ratio of wall and core material 2.1:1 and emulsifier content 3.4%, under the technological conditions encapsulation efficiency can reach 60.79%, average oil load is 21.6%. Microcapsules prepared by emulsion polymerization are selected to have performance characterization and functional verification.Physical and chemical characteristics of the microcapsule were investigated(including moisture content, bulk density, solubility, repose angle, retention ratio and peroxide value of the microcapsules) and then storage stability of the products were researched. The results showed that the pine seed oil microcapsule had a water content of 2.92 ± 0.07%, the bulk density is 0.24 ± 0.04g/cm3, the solubility is 93.19 ±2.41g/100 g and the repose angle is 38.77±0.53°. Under different conditions after 35 days’ storage, the retention rate of pine seed oil microcapsules was decreased, so the microcapsule should be stored at low temperature without light and oxygen. The simulated conditions of gastrointestinal test showed that pine seed oil microcapsules have played an important role for protecting of core material in the stomach and effectively release in the intestine.By the established model of hyperlipidemia in mice, we studied the physiological function of Korean pine seed oil microcapsule. The results showed that Korean pine seed oil microcapsules in losing weight, promoting fat metabolism, relieving lipid peroxidation and enhancing immunity all have significant effects.Therefore, as a functional food therapy health care products, Korean pine seed oil microcapsules has a good prospect of development.
