Malignant tumors are a serious threat to human health,and the incidence rate has been increasing year by year and is becoming younger.Chemotherapy is currently one of the major treatment for malignant tumors.However,chemotherapeutic drugs have shortcomings such as short half-life and high toxicity to healthy tissues.Therefore,researchers have turned their attention to the development of nanocarriers with good biocompatibility and high payloads for the treatment of oncology drugs,and targeted therapy and continuous therapy.The structure and properties of the nanocarriers will affect the antitumor effect of the drug,so it is very important to establish the evaluation system that relationship of the structure-activity of nanocarriers and the antitumor activity.The main results are as follows:1.By controlling the hydrolysis process of proteins,nanospheres and nanotubes of uniform size were constructed based on the self-assembly effect of the product peptides,and the glutaraldehyde was used to impart different flexibility to the particles.At the same time,the length of nanotubes was controlled by the shearing effect.The basic unit of the material was kept unchanged.The above-mentioned strategies were used to synthesis different physical and chemical properties of nanocarriers,which lays a material foundation for the examination and comparison of subsequent biological effects.2.At the level of two-dimensional cells,the correlation between the physical and chemical properties of the particles and the different cellular uptake kinetics was systematically described.In combination with in vitro cell culture techniques,the penetration ability of different nanoparticles and the effect of tumor inhibition were achieved on a three-dimensional level.A systematic study was conducted to determine the advantages of flexible short tubes.3.The metabolism and intratumoral accumulation of different nanoparticles in vivo were elucidated.It was further confirmed the comprehensive advantages of the flexible short tube as an antitumor drug carrier.
